| Purpose: To further discuss the therapeutic effect of "XiaoYongKuiDeKang" decoction on gastric ulcers and its mechanism by observing the effect of "XiaoYongKuiDeKang" decoction on interleukin 6 and interleukin 8 in rats with acetic acid-type gastric ulcer to provide a reliable basis for clinical treatment of acetic acid-type gastric ulcer.Materials and Methods: Forty-six SPF-class healthy Wistar rats(20 rats are male and 20 rats are female)weighing 180±10g were housed in cages and fed for 3days.Then,the rats were randomly divided into 4 groups: the blank group(10),the model Group(12),the omeprazole test group(12)and the XiaoYongKuiDeKang test group(12).The rats with acetic acid-type gastric ulcer were modeled by the glacial acetic acid injection method,and 0.02 ml of 100% glacial acetic acid was injected into the anterior walls of the gastric antrums of rats.When a 3 mm white ulcer was found in the gastric serosa,the modeling was successful,the stomach was returned back into the abdominal cavity and the abdominal cavity was stitched layer by layer.During treatment,4 groups of rats were intragastrically administrated for 14 days.Rats of the blank group and the model group were administrated with 1.25 ml normal saline per100 g body weight every day.Rats of the omeprazole test group were administrated with 1.04 ml omeprazole aqueous solution per 100 g body weight every day.Rats of the XiaoYongKuiDeKang test group were administered with 1.25 ml XiaoYongKuiDeKang decoction per 100 g body weight every day.All rats were anesthetized by injecting 10% chloral hydrate into the abdominal cavity 2 hours after the last administration.After laparotomy,the pylorus and cardia were ligated,and the whole stomach was placed in 10% formaldehyde solution for 20 minutes to fix it.Then,the stomach was cut open,washed with sterile saline,spread,observed and sampled.The effect on the expression of interleukin 8mRNA in gastric tissues of rats was tested by the polymerase chain reaction(PCR)technology.The expression of contents of interleukin 6 in tissues was tested by the enzyme linked immunosorbent assay(ELISA)method.The results were statistically analyzed by variance analysis in the statistical software spss19.0.Results:1.The relative expression and content of interleukin 8 in the model group were increased and are significantly different from those in the normal group(P<0.05).After treatment,the relative expression and contents of interleukin 8 in the XiaoYongKuiDeKang test group and the omeprazole test group were lower than those in the model group(P<0.05),but there was no significant difference between the XiaoYongKuiDeKang test group and the omeprazole test group(P>0.05).2.Compared with the blank group,the content of IL-6 in the model group was significantly increased(P<0.05).Compared with the model group,the content of interleukin 6 in each test group was significantly reduced(P<0.05).There was no significant difference in the content of interleukin 6 between the XiaoYongKuiDeKang test group and the omeprazole test group(P>0.05).Conclusion:1."XiaoYongKuiDeKang" decoction can reduce the relative expression of interleukin8 mRNA in gastric mucosa tissues of rats with acetic acid-type gastric ulcer,and reduce the content of interleukin in tissues,thereby improving the gastric ulcer in rats.2."XiaoYongKuiDeKang" decoction can reduce the content of interleukin 6 in gastric tissues of rats with gastric ulcer,improve the general condition of rats with gastric ulcer,effectively promote ulcer healing and improve the recovery ability of damaged mucosa. |
