Purpose:Basing on the theory of the etiology of traditional Chinese medicine,we made the model of spleen Yin deficiency syndrome rats by classical compound factors modeling method.We detected related indexes of ileal tissue and discussed the mechanism of spleen yin deficiency syndrome from the perspectives of pathological changes and molecular biology.We also used Li Pi Zi Yin method to interpose the rats with spleen yin deficiency syndrome and regarded the results as proofs of evaluation of models.We wished to further enrich the research content of spleen yin deficiency syndrome and provided ideas for subsequent related researches.Material and method:We selected 42 healthy SPF grade SD rats and divided them into control group,model group and treatment group following randomization that according to their gender and weight.Each group had 14 rats and the quantities of males and females were equal.We used classical compound factor molding method--improper diet,excessive labor and traditional Chinese medicine,to make models.There were two phases:firstly,we made spleen Qi deficiency models by means of improper diet and excessive labor,secondly,we made spleen Yin deficiency models with traditional Chinese medicine.Each phase spent 8 days.At the end of each phase,in order to judge the quality of the models,all of them were evaluated by the pattern recognition method of mathematics.We observed changes of ileal tissue and tested the activity or content of GSH-Px,MDA,Na+-K+ATPase,Ca2+-Mg2+ATPase,Complex ? and Complex ? by colorimetry,and tested the content of c AMP,PKA,VIP and ?-EP by ELISA,and tested the expression of protein of c AMP,PKA,VIP and ?-EP by Western Blot.Results:1.The results of swimming endurance,weight and rectal temperature in rats: Compared with the control group,the swimming endurance and weight of the model group decreased significantly(P<0.01),and the rectal temperature increased(P<0.01);Compared with the model group,the swimming endurance and weight of the treatment group increased significantly and the rectal temperature decreased(P<0.01).2.Pathological changes of ileal tissue in rats: In the control group,the ileal mucosa was intact and clear,and the epithelial cells were arranged closely.In the model group,the ileal mucosa became thinner,the villi became shorter or even disappeared,and the epithelial cells appeared necrosis,shedding,hyperemia and inflammatory infiltration;in the treatment group,the ileal mucosa of the rats was still good,the villi were longer,the epithelial cells were arranged more consistently,the necrotic was less,and the inflammatory infiltration was less,too.3.The results of biochemical indexes in each group: Compared with the control group,the activity of GSH-Px in the model group was significantly decreased(P<0.01),the quantity of MDA was significantly increased(P<0.01),the activity of Na+-K+ATPase and Ca2+-Mg2+ATPase were significantly decreased(P<0.01),the activity of Complex I and Complex ? were significantly decreased(P<0.01).Compared with the model group,the activity of GSH-Px in the treatment group was increased(P<0.01),the quantity of MDA was decreased(P<0.01),the activity of Na+-K+ATPase and Ca2+-Mg2+ATPase were significantly increased(P<0.01),the activity of Complex I and Complex ? were significantly increased(P<0.05)(P<0.01).4.The quantities of c AMP,PKA,VIP and ?-EP in ileal tissue were tested by ELISA:Compared with the control group,the quantities of c AMP,PKA and VIP in the model group were significantly decreased(P<0.01),and the quantity of ?-EP was significantly increased(P<0.01).Compared with the model group,the quantities of c AMP,PKA and VIP in the treatment group increased significantly(P<0.01),and the quantity of ?-EP decreased significantly(P<0.01).5.The expression of c AMP,PKA,VIP and ?-EP in ileal tissue were tested by Western blot:Compared with the control group,the expression of c AMP,PKA and VIP in the model group were significantly decreased(P<0.01),and the expression of ?-EP was significantly increased.The expression of c AMP,PKA and VIP in the treatment group were significantly increased(P<0.01),and the expression of ?-EP was significantly decreased(P<0.01).Conclusion:1.The energy metabolism disorders when spleen Yin deficiency syndrome occurs,it is related to the imbalance of oxidative/antioxidative system,the destruction of cells' structure and function,and the retard of oxidative phosphorylation process.2.The digestive system function is weakened when spleen Yin deficiency syndrome occurs,it is related to the decrease of intestinal water secretion and the inhibition of intestinal movement caused by decrease of signal transduction of the brain-gut axis and imbalance of intestinal hormone.
