Purpose:By analyzing the types and contents of active ingredients in ice grapes by high performance liquid chromatography,the edible value and medicinal value of ice grapes can be better defined,which lays a good theoretical foundation for the development of ice grape health food in the future.It is mainly divided into two parts: the first part is to determine the content of gallic acid,catechin,epicatechin,chlorogenic acid,caffeic acid,ferulic acid and rutin in ice grapes by different content determination methods;the second part is The data and methods measured by the above experiments were statistically analyzed using SPSS19.0.Material and method:Fixed-wavelength isocratic elution method using orthogonal analysis,preferably experimental conditions,and using Diamosil C18 column,diameter 5 ?m(4.6× 150mm);column temperature: room temperature;epicatechin,catechin,ferulic acid The mobile phase of chlorogenic acid and caffeic acid is acetonitrile-0.05% phosphoric acid(20:80,V/V),and the mobile phase of gallic acid and rutin is methanol-0.5%phosphoric acid(15:85,V/V);1.0 m L/min;injection volume 10 u L;epicatechin,catechin detection wavelength is 280 nm,ferulic acid,chlorogenic acid,caffeic acid detection wavelength is 325 nm,gallic acid,rutin detection wavelength 270 nm,356 nm.A fixed-wavelength gradient elution method and a multi-assay method using an Agilent C18 column(4.6× 250 mm,5 ?m),mobile phase acetonitrile(A)-acetic acid: water = 2:98(B),gradient elution [0 min,AB(6:94);14 min,AB(12:88);30 min,AB(20:80);40min,AB(40:60)],flow rate 1.0 m L/min,detection The wavelength is 280 nm and the injection volume is 10 ?L.The wavelength-switched gradient elution method uses an Agilent C18 column(4.6×250 mm,5 ?m),and the mobile phase is acetonitrile(A)-acetic acid: water = 2:98(B),and the injection volume is 10 ?L.: room temperature,gradient elution [0 min,AB(6:94);14 min,AB(12:88);30 min,AB(20:80);40 min,AB(40:60)].See Table 1 forwavelength switching.(?)At the same time,SPSS19.0 statistical analysis software was used to statistically analyze the data of the components in the ice grapes measured before,and compare the differences between the methods.Results:The types and amounts of the components in the ice grapes measured by the fixed wavelength isocratic elution method are as follows: gallic acid,catechin,chlorogenic acid,caffeic acid,epicatechin,ferulic acid and,rutin are respectively 13550 ?g/g,78110?g/g,262.0 ?g/g,895.8 ?g/g,31500 ?g/g,80.88 ?g/g,764.5 ?g/g.The types and amounts of the components in the ice grapes measured by the fixed wavelength gradient elution method are as follows: gallic acid,catechin,chlorogenic acid,caffeic acid,epicatechin,ferulic acid and rutin are respectively: 8409 ?g/g,50090?g/g,59.67 ?g/g,132.0 ?g/g,8014 ?g/g,70.20 ?g/g,653.5 ?g/g.The types and amounts of the components in the ice grapes measured by the wavelength switching gradient elution method are as follows: gallic acid,catechin,chlorogenic acid,caffeic acid,epicatechin,ferulic acid and rutin are respectively: 8604 ?g/g,55700 ?g/g,159.0 ?g/g,528.0 ?g/g,16130 ?g/g,71.37 ?g/g,676.6 ?g/g.The types and amounts of the components in the ice grapes measured by the multi-evaluation method are as follows: gallic acid,catechin,chlorogenic acid,caffeic acid,epicatechin,ferulic acid and rutin are respectively: 8469 ?g/g,50775 ?g/g,5089?g/g,134.9 ?g/g,8128 ?g/g,69.98 ?g/g,650.5 ?g/g.The statistical analysis part uses the covariance analysis method,the clustering method in the cluster analysis method,the rapid clustering method and the discriminant analysis method to analyze the obtained data,and obtains the corresponding statistical analysis conclusion: one measurement multi-evaluation method There is no significant difference from the external standard method;the measured seven compounds are divided into three categories according to the content,and the measured content can be quickly referred to the type and name of the compound by substituting the measured content into the discriminant equation.Conclusion:1.Ice grapes contain 7 different compounds,namely gallic acid,catechin,chlorogenic acid,caffeic acid,epicatechin,ferulic acid and rutin.2.There are basically no influences on the types of determination of the content of the seven compounds by different content determination methods.3.Statistical analysis proves that the one-test multi-evaluation method is applicable to the determination of seven different compounds in ice grapes;it is applicable in the quality evaluation of seven different components of ice grapes.
