Purpose:To evaluate the ability of Liao gaoben to regulate such indexes as lipid,C-reactive protein and blood flow in atherosclerosis model rabbits.To observe the effect of Liao gaoben on the total carotid artery in rabbits and explore the possible mechanism of its antiatherosclerosis.Material and method:After 1 week of adaptive breeding of male Japanese big ear white rabbits,they were randomly divided into blank groups(10)and model groups.With the exception of the blank group,the model group used immune injury method,balloon surgery method,and high-fat feed feeding method to complete the atherosclerosis animal model.Afterwards,the molded white rabbits were randomly divided into model groups,Liao Liben clinical 4x equivalent dose group(1.88 g crude medicine/kg),Liao Liben clinical 2x equivalent dose group(0.94 g crude medicine/kg),and Chinese medicine positive control.Lipoid Kang group,In addition,there are 5 groups of white rabbits in the blank group,each with 10.On the second day after the moulding operation,gastric administration began.The blank group and the model group gave equal amounts of distilled water once a day for 4 consecutive weeks.According to the conditions of the White rabbits,the remaining groups were given the corresponding dose of drug enema,and the clinical equivalent dose group of Liao Liben was given 1.88 g raw medicine/kg to the White rabbits,and the clinical equivalent dose group of Liao Liben was given to the White rabbits.0.94 g raw medicine/kg,Zhongyang Formation: Based on the model group,Shizeikang irrigation stomach.1 per day for 4 consecutive weeks.After the treatment,the male white rabbit is anaesthetized for abdominal aorta blood,Collection of blood for triglyceride(TG)determination,total cholesterol(TC)determination,high-density lipoprotein(HDL)determination,low-density lipoprotein(LDL)determination,high-density lipoprotein(HDL-2),pre-?1 high-density lipoprotein(pre?1-HDL),C Reaction protein;Collection of whole blood and plasma for hemorheology test;The pathological morphology of the total carotid artery endothelium was observed by HE staining.Results: 1.Compared with white rabbits in the model group,TG,TC,and LDL all increased,with significant differences(P<0.05).Combined with the pathological morphology of white rabbits in the HE staining model group,the results showed that hyperlipidemia and atherosclerosis were successfully established.model.However,there was no significant change in pre?1-HDL,indicating that the molding method had no significant effect on pre?1-HDL.2.Compared with the model group,the 2 times clinical equivalent dose group(0.94 g crude medicine/kg)of Liaoji was able to significantly reduce the LDL level(P<0.05),and had a decreasing trend for TC and TG levels(P>0.05),but there was no significant difference;Compared with the model group,Liao Liben clinical 4 times equivalent dose group(1.88 g raw medicine/kg)was able to significantly reduce TC and LDL levels(P<0.05),and there was a decreasing trend for TG levels(P>0.05),but there was no significant difference;Compared with the blank group,HDL and HDL2 were significantly increased and had significant differences(P<0.05).Compared with the model group,the content of HDL2 increased,but there was no significant difference(P > 0.05),and there was no change in HDL content.3.Compared with the blank group,the C-reactive protein in the serum was significantly higher and had significant difference(P<0.05).Compared with the model group,the contents of C-reactive protein decreased significantly(P<0.05).4.Compared with the blank group,the high,moderate,low and plasma viscosity and erythrocyte aggregation index of the whole blood increased significantly(P < 0.05).Compared with the model group,the two clinical equivalent dose groups of Liao Sakamoto and four clinical equivalent dose groups of Liao Sakamoto,the total blood high cut,medium cut,low cut,plasma viscosity,red blood cell aggregation index,and significant decrease,(P< 0.05).5.The thickness of the carotid artery in blank group was uniform,the inner membrane was smooth,the structure was complete,and the endothelial cells had no damage.There was no obvious lesions.The thickness of arterial wall in the model group was different and uneven.Obvious atherosclerotic plaques were seen.Endothelium cells were damaged,detached,inner membrane hyperplasia,and a large number of foam cells and a small amount of cell fragments were seen under the inner membrane.A large number of fibrous plaque hyperplasia and endothelial cell exfoliation were observed in the lipids group.A large number of irregular proliferation of fiber plaques and degenerative foam cells were observed in the 2 times equivalent dose group.A small amount of fibrous plaque hyperplasia was observed in the 4 times equivalent dose group of gaoben clinic,and the accumulation of foam cells under the inner membrane decreased significantly.Conclusion: Liao gaoben has a clear role in regulating blood lipids;Liao Sakamoto has a clear effect on the reduction of C reaction protein in AS white rabbits;Liao Sakamoto has a clear effect on the improvement of hemodynamics in AS white rabbits;Liao gaoben has the effect of improving the pathological morphology of AS white rabbits.
