Purpose:Observe the effect of Rhizoma Chuanxiong and peony on the nerve growth factor(NGF),brain-derived neurotrophic factor(BDNF),platelet-derived growth factor(PDGF),and transforming growth factor(TGF)after cerebral ischemia-reperfusion;To explore the neuroprotective effect of Rhizoma Chuanxiong and Radix Paeoniae on cerebral ischemic injury;It provides a theoretical basis for the treatment of ischemic stroke.Material and method: The experimental rats(SPF grade,male SD rats,body weight 220-280g)were 210 animals,free movement,drinking water,feeding.The body weight of each rat was weighed,and the rats were anesthetized with 3% chloral hydrate for intraperitoneal injection.The rat model of focal cerebral ischemia and reperfusion was prepared by a Longa suture method.The rat model was inserted into the line from the internal carotid artery,length of insertion line18.5 ± 0.5 mm,2 h after the modeling,the inserted peg was pulled out 1.0 cm,and the excess part of the skin was cut off.Rat neurological function was then scored according to the Zea Longa and Bederson 5-point scale.Scores were scored at 1-3.Not included in the criteria: score 0 points;score 4 points;subarachnoid hemorrhage.Rats that died during removal of the model or did not reach the brain were sacrificed randomly in the deficient group.The experimental rats were randomly divided into 8 groups: blank group(normal rats,without any treatment),sham operation group(only skin was incised,and the right common carotid artery was dissected and sutured,and no other treatment was performed).model group,the high-dose group,middle-dose group,low-dose group,ginkgo leaf group,and nimodipine group were found in each of the18 groups.After intragastric administration,the rats in the blank group,sham operation group,and model group were given an equal volume of distilled water according to the weight of the rats,and the remaining groups were given the corresponding dose of the therapeutic drug once a day.After 2 hours on the 3d,7d,and 14 d administration groups,6 rats in each group were sacrificed.The brains of the rats were taken out,and then the olfactory bulb,cerebellum,brain stem,frontal pole,and normal side brain tissue were cut off,and the right ischemic brain tissue was preserved.The levels of PDGF and NGF in ischemic brain tissue were detected by ELISA.The expression of TGF mRNA and BDNF mRNA in ischemic brain tissue were detected by Real-time PCR.Results:1.The content of NGF and PDGF in the brain tissue of rats with ischemia-reperfusion was detected by ELISA after 3d,7d and 14 d.Between the blank and sham-operated groups,the content of NGF and PDGF in the brain did not change significantly(P>0.05).The expression of NGF and PDGF in the brain of the other groups increased and was significantly higher than that of the blank group.The difference was statistically significant(P<0.05).The contents of NGF and PDGF in ischemic brain tissue in sham-operated rats were significantly lower than those in model group(P<0.05);the brain tissues of low-dose,middle-dose,high-dose,nimodipine,and ginkgo leaf groups of rhizoma chuanxiong and peony.The expression of NGF and PDGF was significantly higher in the model group than in the model group(P<0.05).2.RT-PCR method was used to detect the expression of TGF mRNA and BDNF mRNA in brain tissue of rats after cerebral ischemia reperfusion for 3d,7d and 14 d.There was no significant difference in mRNA expression of TGF-? and BDNF between the blank group and the sham group(P>0.05).The mRNA expression levels of TGF-? and BDNF in the other groups were significantly higher than those in the blank group,with significant differences(P<0.05).Compared with the model group,the expression of TGF mRNA and BDNF mRNA in the sham operation group was decreased(P<0.05);the low-dose group,middle-dose group,high-dose group,nimodipine group,ginkgo leaf group,and ischemic brain tissue of rhizoma chuanxiong and peony.The expression of TGF mRNA and BDNF mRNA increased significantly(P<0.05).Conclusion:1.Increased leves of NGF and PDGF are beneficial to the recovery of damaged brain tissue and brain nerves.Ligusticum chuanxiong and Radix Paeoniae Rubra couldpromote the increase of NGF and PDGF contents,suggesting that chuanxiong and Radix Paeoniae Rubra may play a neuroprotective role by increasing the content of NGF and PDGF.2.Increased expression of BDNF and TGF is beneficial to the recovery of damaged brain tissue and brain nerves.Ligusticum chuanxiong and Radix Paeoniae radix can upregulate the expression of BDNF mRNA and TGF mRNA,suggesting that chuanxiong and Radix Paeoniae Rubra may increase the expression of BDNF mRNA and TGF mRNA.Neuroprotective effect.
