Purpose:The effect of excessive fatigue on the heart,vascular endothelium and blood lipid of Bama minitype pigs with spleen qi deficiency syndrome was investigated through overexercise.Material and method:The 6 Guangxi Bama miniature pigs were randomly divided into two groups,each group has 3,the control group only treated with high salt and high fat diet,the overexercise group with high salt and high fat diet and given excessive exercise intervention,daily 9:00,15:00 feed,daily evening training run time.he experiment began zeroth weeks of overexercise group pig adaptability training for 1 weeks running,starting from second weeks into the campaign over running the training phase,the pig running pace of neat uniform,not stumble and fall phenomenon is considered to be the most suitable running speed,and the pig continued appearing in the process of running and refused to roar continue running,fall,lie can not afford,takes a breath that has reached the limit of the pig run,Total running for 16 week,respectively in the zeroth week,4 week,8 week,12 week and 16 week were observed in two groups of symptoms of pig spleen deficiency syndrome and detection of vascular endothelial growth factor and blood lipid index changes,the end of the sixteenth week after the experiment by echocardiography to detect the structure and function of the pig heart,after the end of the experiment pig and executed at the apical tissue,coronary artery,thigh muscle,abdominal fat and left ventricular Endothelin-1(ET-1),Nitric oxide(NO)and Vascular endothelial growth factor(VEGF)were detected by HE staining and structure of pig aorta and ventricular muscle were observed.Results:1?Establish the spleen deficiency model:Through the comparison of two groups of pig experimental data,found that the stool and waiting to be fed for overexercise behavior score group was higher than control group and there was significant difference;eces scratches,fur,mouth color though there is no statistical significance,but also the eoverexercise group mean higher than the control group.2?Echocardiography:Overexercise group of LVEDD was higher than that of control group(44.43±1.88 VS.39.27±0.64)mm,P<0.05;overexercise group of EDV was higher than that of control group(89.90±8.81 VS.67.50±2.45)ml,P<0.05;overexercise group of SV was higher than that of control group(71.5±8.15 VS.54.80±1.50)ml,P<0.05;overexercise group of LVPWS was higher than that of control group(14.53±0.06 VS.13.30±0.00)mm,P<0.05;Overexercise group of LVPWD was higher than that of control group(9.13±0.98 VS.8.37±1.22)mm,P>0.05;overexercise group of ESV was higher than that of control group(18.4±4.86 VS.12.70±0.95)ml,overexercise group of LVESD was higher than that of control group(23.03±2.45 VS.20.07±0.61)mm,P>0.05;The HR overexercise group was lower than the control group(93.00±12.29 VS.145.33±1.53)times/min,P<0.05;CO overexercise group was lower than the control group(6582.83±72.36 overexercise group was lower than the control group(48.13±5.11 VS.49.57± 0.93)%,P>0.05;E peak flow overexercise group was lower than the control group(0.83±0.06 VS.0.91±0.15)m/s,P>0.05;A peak flow was lower than the control group(0.58±0.09 VS.0.59± 0.10)m/s,P>0.05.3?Vascular endothelial factor:(1)In the organization:NO in the coronary artery tissues was all over the overexercise group,which was smaller than that in the control group(0.12±0.01 VS.0.21±0.04)ng/L,P<0.05;VEGF in the apex tissue hyperkinetic group was smaller than that in the control group(0.18±0.01 VS.0.22±0.01)ng/L,P<0.05.(2)In serum:VEGF eoverexercise group compared with the control group P<0.01,VEGF in the twelfth and sixteenth weeks were all over the exercise group lower than the control group(0.10±0.01 VS.0.20±0.04?0.13±0.01 VS.0.28±0.06)ng/L,P<0.05.ET-1 in twelfth weeks when overexercise group than the control group(0.10±0.01 VS.0.14±0.00)ng/L,P<0.05;NO in sixteenth weeks when excessive exercise group than the control group(0.14±0.01 VS.0.24±0.05)ng/L,P<0.05;but in other tissues,serum ET-1,NO,VEGF between the two groups had no significant difference.4?Aortic and ventricular muscle structure:The aorta and ventricular muscle of piglets were detected by HE staining,At sixteenth weeks,the morphology of aortic smooth muscle cells in the control group was spindle shaped and arranged in a slightly uneven pattern.The nuclei were spindle shaped and arranged slightly irregularly,with a slight change in size.In the overactive group,the morphology of vascular smooth muscle cells was obviously changed.The gap between some cells was enlarged and irregular.The nuclear morphology was cylindrical and arranged in disorder,and atherosclerosis in the subintimal layer was seen.The control group at sixteenth weeks the pig left ventricular myocardial cells increased slightly,slightly arranged muscle disorders,muscle interval slightly widened,no intercalated disc and striated muscle,nuclear morphology of spindle shaped,arranged slightly irregular,nuclear size is approximately equal;excessive exercise group cardiomyocytehypertrophy,disordered muscle septum,the muscle bundle the hyperplasia of fibrous tissue,muscle fiber fracture,no intercalated disc and striated muscle,the nucleus was round in shape,irregular arrangement,nuclear size.5?Blood lipid:there was no statistical difference between the two groups of TG,CHOL and LDL-C between the two groups had no significant difference,P>0.05;HDL-C at sixteenth weeks when overexercise group than the control group(0.64±0.08 VS.0.94±0.10)mmol/L,P< 0.05;and LDL-C 0 week,4 week,8 week,12 week and 16 week the two groups were compared for excessive exercise group was higher than control group(1.32±0.14 VS.1.12±0.17,1.36±0.11 VS.1.12±0.17,1.43±0.2 VS.1.19± 0.18,1.56±0.19 VS.1.36±0.28,1.20 ±0.16 VS.0.83±0.72)mmol/L,P>0.05.Conclusion:1?The after overexercise,pig excrement was loose,waiting to be fed more indifferent,stool scratches more clear,more dry hair gloss,color and more bruising,namely excessive exercise group of spleen qi deficiency symptoms significantly pig.2?The after overexercise,the heart wall thickness of piglets is thickened,the volume of ventricle increases,and the ejection fraction decreases.The more significant the syndrome of spleen qi deficiency syndrome is,the more significant the change of heart structure and function of the pig.3?The after overexercise,the content of VEGF in serum,NO in coronary artery and the VEGF in apical tissue were decreased in serum.4?The after overexercise,the morphological structure of vascular smooth muscle cells and cell nuclei of the aorta of the pig was not as good as that of the control group,atherosclerotic changes in the underlayer of the endomintima;The morphological structure of left ventricular myocytes,muscle fibers and nuclei were not as good as that of the control group;It can indicate that the more obvious symptoms of spleen qi deficiency syndrome will aggravate the damage of the vascular and ventricular muscle structure of the piglets.5?The after overexercise,There was no statistical difference between the two groups of TG,CHOL,HDL-C and LDL-C,P>0.05.
