Objective:The aim of this research is to elucidate the influence of BuqiHuoxueTongluo Formula on induced IPF rat model through the aspect of histopathology and PI3K/AKT signal pathway,and on this basement,further study the effect on pulmonary function test and the secretion of AECII.Methods:Experiment 1:80 male Wistar rats were divided into four groups according to the random number table,20 cases in each group:control group,BLM group,BLM +Pred group,BLM + BHTF group.3mg/kg BLM was injected intratracheally to induce IPF model.The next day after establishing the model,rats of BLM + Pred group were given prednisone 3.6mg/kg/d by intragastric gavage,BLM + BHTF group was treated with a decoction of BuqiHuoxueTongluo Formula 16.5 g/kg/d by intragastric gavage,and control group and BLM group were given the same volume of normal saline per day.On the 14th and 28th day from inducing model,respectively,half of rats were randomly sacrificed from each group.hematoxylin-eosin and Masson's trichrome staining was performed to observe the histopathologic changes and calculate the IOD.Immunohistochemistry staining was performed to observe the position and semi-quantitative measurement of TGF-?1 and ?-SMA in lung tissue.The expression of PI3K/AKT signal pathway was measured by the methods of RT-PCR and Western Blot.Experiment 2:The protocol of experiment was the same with Experiment 1.On the 14th and 28th day from inducing model,before being sacrificed,rats underwent the pulmonary function test under anesthesia.Rats were tracheostomized and the Anires 2005 animal ventilator system and graphed the pressure-volume loop,calculated the area of hysteresis.TPL was measured by colorimetric method in BALF.The ultra microstructure was observed by TEM.Results:Experiment 1:Compared with other BLM-induced groups,there were lightly inflammation changes and fibrotic deposition in BHTF Group.The pathological score and IOD had statistically significant difference.The positive area and IOD of TGF-?1 and ?-SMA had obvious difference contrast with BLM and BLM+Pred Group.To the expression of PI3K/AKT mRNA and protein,the BHTF Group had statistically significant difference with others.Experiment 2:The ventilation function of BHTF Group was significantly improved compared with BLM and BLM+Pred group,the compliance of lung tissue and the shape of P-V Loop was maintained better.BHTF group had a more preferable lung capacity than BLM and Pred Group with a wider area between the two limbs,which was defined as hysteresis.TPL in BALF of BHTF Group had statistically significant difference compared with BLM and BLM+Pred Group.The BHTF Group had more lamellar bodies and the structure was relatively normal than BLM group.Conclusion:Through the PI3K/AKT signal pathway,BuqiHuoxueTongluo Formula could ameliorate the deterioration of histopathology and compliance of lung tissue,further speaking,it could maintain the normal shape of P-V Loop and hysteresis,improve the work of breath.The micro foundation of these was the maintain of secretion function and normal ultra micro structure.
