Objective:Clinically,high levels of HBV DNA replication,ALT and AST in serum within the normal range,and liver histological examination showed no lesion or mild lesion,are commonly seen among chronic hepatitis B virus carriers.They are highly infectious,and may suffer from liver cirrhosis or hepatocellular carcinoma as the disease progress secretly.Chronic HBV carriers are in the immune tolerance stage,and there is no ideal treatment for such patients in modern medicine.China is a large country with HBV infection,with a large number of chronic HBV carriers and a heavy disease burden,so it is particularly important to solve this problem.Previous study have found that the Fuzheng Qudu formula can inhibit HBV replication in chronic HBV carriers and up-regulate the expression of IFN-?.Based on the above research,we designed the following experiment,using the Fuzheng Qudu formula to intervene the HBV transgenic mice.By observing ALT,AST,HBV DNA,spleen index,IFN-?,IL-10 and hepatic pathology,we want to discuss its influence on liver cells and replication of HBV,and research its immunomodulatory effects on the transgenic mice.Method:Randomly divided 30 cases HBV transgenic mice into Chinese medicine group,western medicine group and model group,10 cases corresponding normal mice were set as blank group.Chinese medicine group was given the Fuzheng Qudu fromula,the western medicine group was given Adefovir Dipivoxil,the rest two groups were given normal saline.After successive administration for 12 weeks,the animals were sacrificed and the samples were obtained.We use automatic biochemical analyzer to test the level of ALT and AST in the serum,use the quantitative polymerase chain reaction to detect HBV DNA,use ELISA method to measure IFN-? and IL-10 in liver tissue,and HE staining to observe hepatic pathology.Result:1.The average level of ALT and AST:The level of ALT in the model group is higher than the rest three groups,and the differences all have statistical significance(P>0.05).The level of ALT among the Chinese medicine group,the western medicine group and the blank group have no difference(P>0.05).The average level of AST has no difference among the four groups(P>0.05).2.HBV DNA relative expression:The relative expression of HBV DNA has no difference between the western medicine group and the Chinese medicine group(P>0.05).3.The average level of spleen index:The average level of spleen index in the blank group is higher than the rest three groups,which have statistical significance(P>0.05),and the differences among the model group,the Chinese medicine group and the western medicine group have no statistical significance(P>0.05),while the average level of spleen index in the Chinese medicine group and the western medicine group are higher than that in the blank group.4.The average level of IFN-?:The average level of IFN-? in the western medicine group is higher than the rest three groups,and the differences all have statistical significance(P<0.05).The difference of the average level of IFN-? between the model group and the blank group has no statistical significance(P>0.05).And the difference of the average level of IFN-y between the model group and the Chinese medicine group has no statistical significance(P>0.05)either.5.The average level of IL-10:The average level of IL-10 in the blank group is lower than the rest three groups,and the differences all have statistical significance(P<0.05).The average level of IL-10 in the model group is lower than that in the the western medicine group,and the difference has statistical significance(P<0.01).The difference of the average level of IL-10 between the model group and the Chinese medicine group has no statistical significance(P>0.05).6.Hepatic pathology:Hepatocytes are poorly organized in the model group,hyaline degeneration and vacuole like denaturation can be observed,while there is no significant lymphocyte infiltration,portal area fibrosis is not observed either.Hepatocytes of the rest three groups are well organized without obvious pathological changes.Conclusion:1.The Fuzheng Qudu formula can alleviate the damage of hepatocyte in transgenic mice,and it may improve the prognosis of chronic HBV carriers.2.Fuzheng Qudu formula cannot affect the expression of immune factors(IFN-?,IL-10,et al)in the transgenic mice in the short term.3.The Fuzheng Qudu formula may have the effect of inhibiting HBV replication in chronic HBV carrying mice.
