Psoriasis is a multi-gene-mediated immune-related disease characterized by erythema and scale.Because of its high incidence and recurrence rate,quite a lot of patients suffer from this disease.It also imposes an adverse impact on society..Since the Ming and Qing Dynasties,the "blood-score treatment“of psoriasis has been widely accepted as a stable and mature theory in the clinic.The"Diagnostic Efficacy Standards for TCM Syndrome" and "Differential Diagnosis of TCM Symptoms"divide psoriasis into blood-heat type,blood-stasis type,and blood-dry type.The differences between these syndrome types of psoriasis are not only manifested in the lesions,but also in the blood and skin tissue,such as T lymphocytes,cytokines,genomics,proteomics,neuroendocrine immune network,and vascular endothelial growth factor.The current study found that there are significant differences of microRNAs in various tissues and developmental stages of the disease.These microRNAs participate in a variety of biological processes,and are closely related to a variety of physiological and pathological processes,and can well reflect the interaction of genes and the environment.The study found that more than 250 microRNAs are abnormally expressed in blood and tissue of psoriasis.For example,miR-203 is highly expressed in lesions of psoriasis patients,causing proliferation and differentiation of keratinocytes by inhibiting the expression of SOCS;likewise,miR-146a is highly expressed in peripheral blood of patients with psoriasis,which impairs the body's natural immunity and promotes inflammatory response through inhibiting the expression of TARF6 and IRAKI;similarly,miR-125b is lowly expressed in blood of psoriasis patients,regulating the proliferation and differentiation of keratinocytes.The aforementioned examples indicate that microRNA plays an important role in the pathogenesis and progression of psoriasis.In addition,microRNA is also an important part that can not be ignored in the research of disease TCM syndrome type,due to its obvious differences in blood or skin tissue of patients with different syndromes types of coronary heart disease and tumor.There are some unique features of the treatment of psoriasis in Traditional Chinese Medicine.After years of clinical verification,the therapy from Traditional Chinese Medicine has become an important method for the treatment of psoriasis.Syndrome types are distinguished by lesions and internal factors,which means that it is affected by both genes and factors such as environment and living habits.The etiology of psoriasis is complex and the clinical manifestations are diverse.Despite the experience and guidance of the famous experts,not all clinicians can correctly grasp the syndrome differentiation and treatment of psoriasis,resulting in uneven clinical efficacy.Therefore,rigorous experimental indicators can contribute to determining the clinical classification of psoriasis accurately.To this end,we aim to study the differences in microRNAs between different syndrome types,to further reveal the psoriasis blood treatment and the nature of each psoriasis syndrome,and to provide a biological basis for the "blood treatment" of psoriasis.Objective:To investigate the differential expression of microRNA in peripheral blood of patients with different syndromes type of psoriasis,and the clinical significance of specific miRNAs in the pathogenesis of psoriasis.Methods:MicroRNA-chip technology was used to detect some different miRNAs;target gene prediction and target gene function enrichment analysis were performed on different miRNAs.Results:1.Comparison of miRNAs between groups:There were 51miRNAs in PBMCs of normal people and patients with blood-heat psoriasis.Among them,10 were up-regulated,such as miR-125b-5p,and 41 were down-regulated,such as miR-31-3p.There were 217 miRNAs in PBMCs of normal people and blood-stasis psoriasis patients.Among them,65 were up-regulated 5 and 152 were down-regulated?such as miR-125b-5p.There were 180miRNAs in PBMCs of patients with blood-heat and blood-stasis psoriasis.Among them,121 were up-regulated?such as miR-200b-3p,and 59 were down-regulated 5 such as miR-155-5p and miR-125b-5p.2.Target gene prediction and biological analysis of differentially expressed miRNAs.By predicting the target genes of differential miRNAs and performing GO analysis on the predicted target genes(see Figure 3),the results indicate that the biological processes involved in the target genes are GO:0048518positive regulation of biological process;GO:0035556intracellular signal transduction;GO;0023051 regulation of signaling;GO:0010646 regulation of cell communication,etc.,indicating that these biological processes are involved in the differential expression of different syndromes of psoriasis.By performing a pathway analysis based on the KEGG database of target genes of differentially expressed miRNAs,a number of significantly enriched signaling pathways were obtained.The results are as follows(see Figure 4),indicating that these pathways are differentially expressed in different syndromes of psoriasis.Important role.Conclusion:There are significant differences in miRNAs between normal people and patients with blood-heat psoriasis,normal people and patients with blood-stasis psoriasis,patients with blood-heated psoriasis and patients with blood-stasis psoriasis.miRNAs may play an important role in the development of different syndromes of psoriasis.
