Objective:(l)Observing the effect of moxibustion on the cachexia process in gastric cancer rats.(2)The therapeutic effect of moxibustion on skeletal muscle consumption in rats with gastric cancer cachexia was studied from the changes of muscle weight and myocyte morphology and myocyte apoptosis.(3)The effect of moxibustion on the proinflammatory factors TNF-? and IL-6,ubiquitin ligase MAFbx and MuRF-1 in the gastrocnemius muscle,and elucidated the possible mechanism of moxibustion for the treatment of gastric cancer cachexia.Methods:Fifty healthy SPF SD rats were randomly selected from the blank group(group A)according to the random number table method.Twelve patients were randomly selected to simulate the tumor transplantation process.After successful,10 patients were randomly assigned to the sham operation group(group B).The remaining 28 patients were treated with gastric adhesion of Walker 256 tumor tissue.Seven days later,3 randomly selected to verify the success of gastric cancer modeling,the remaining 25 observed changes in body mass,and reference to the classification of human cachexia status,when the body mass of gastric cancer rats was reduced to 95%of the tumors were randomly divided into the model group(group C)and the moxibustion group(group D),with 10 rats in each group.From the first day after the enrollment,the moxibustion group alternated the mild moxibustion 1 in the next day.the middle spleen,Guanyuan,Zusanli 2 spleen and stomach spleen.At the same time,the blank group,the sham operation group and the model group were accompanied by the grasping and binding.Each group was intervened for 20 min each time,once a day for 14 days.The body weight,food intake and behavioral changes of rats-in each group were observed.After the end of the treatment,the animals were sacrificed,the mass of the organ was measured,the muscle weight of the bilateral gastrocnemius muscle was measured,the morphological changes of the gastrocnemius muscle were measured by HE staining(the transverse diaineter of the muscle cells were measured).and the gastrocnemius muscle was observed by the TUNEL method.Apoptosis and enzyme-linked immunosorbent assay(ELISA)were used to detect the contents of TNF-? and IL-6 in the gastrocnemius muscle.The expression of MAFbx and MuRF-1 in the gastrocnemius muscle was detected by Western Blot.Result:(1)There were no statistically significant differences in dietary intake and body mass between the two groups(P>0.05).Compared with the blank group,the feeding amount of the model rats decreased significantly(P<0.01)from the 12th day of modeling.The body mass of the modeling animals decreased,with significant difference(P<0.05).During the treatment period of moxibustion group,compared with the blank group,the amount of food intake of model group rats continued to decrease,and the difference was significant(P<0.01).At the 6th day of moxibustion,compared with the model group,the food intake of moxibustion group was significantly higher than that of the model group(P<0.05),and after the 14th day of moxibustion,the food intake of moxibustion group was significantly higher than that of the model group(P<0.01).Similarly,during the moxibustion period,the body mass of the moxibustion group began to pick up,and the body mass of the moxibustion group began to be higher than that of the model group(P<0.05).Compared with the model group,the body mass of moxibustion group rats increased significantly the end of moxibustion treatment(P<0.01).(2)Compared with the blank group,the body mass of model group decreased(P<0.01),the mass of gastrocnemius muscle decreased(P<0.01),the transverse diameter of gastrocnemius muscle fiber cells decreased(P<0.01),and the apoptosis of gastrocnemius muscle fiber cells increased(P<0.01).After 14 days of moxibustion treatment,compared with the model group,the body mass of the moxibustion group increased(P<0.01),the mass of the gastrocnics muscle increased(P<0.01),the transverse diameter of the gastrocnics muscle fiber increased(P<0.01),and the apoptosis of the gastrocnics muscle fiber decreased(P<0.05).(3)In addition,compared with the blank group of rats,the level of TNF-,i1-6 cytokines in the gastrocnemius muscle tissue of model group increased(P<0.01),MAFbx protein expression level increased(P<0.01)and murf-1 protein expression level increased(P<0.05).It suggests a cachexia state.After moxibustion treatment for 14 days,the levels of TNF-,il-6 cytokines and MAFbx and murf-1 proteins in the gastrocnella muscle tissues of the moxibustion group were decreased(P<0.05)and the levels of TNF-and il-6 cytokines were decreased(P<0.05).Conclusion:(1)Moxibustion has a therapeutic effect on skeletal muscle depletion in rats with gastric cancer cachexia,which is characterized by increasing skeletal muscle weight,improving pathological changes and reducing myocyte apoptosis.(2)The therapeutic effect of moxibustion on skeletal muscle depletion in rats with gastric cancer cachexia and the reduction of the expression of proinflammatory cytokines TNF-?,IL-6 and ubiquitin ligase MAFbx and MuRF-1 in muscle tissue to inhibit myositis response and reduce Related to protein degradation.
