Objective To observe the effect of Xintong Fang on intracellular NCX mRNA expression and the free calcium ion concentration(Ca2+)in cardiomyocytes of acute myocardial infarction?AMI?rats,and to explore the significance of calcium homeostasis in AMI and the therapeutic mechanism of Xintong Fang.Methods Fifty SD rats were divided into the sham operation group,model group,the hi gh-dose group of Xintong Fang,the low-dose group of Xintong Fang,dilti azem group.The rat model of acute myocardial infarction was established by ligating the left anterior descending branch of the coronary artery in rats,and the Ca2+in myo cardium was detected by laser scanning copolymerization microscope,and the determina tion of myocardial tissue by HE staining,and the expression of NCX mRNA in myocardiu m was measured by RT-PCR.Results 1.Compared with the sham operation group,the NCX mRNA expression decreased a nd the calcium ion concentration increased in the model group?P<0.01?.2.Compared with model group,NCX mRNA expression,calcium ion concentration,and myoca rdial infarct size were decreased in each drug group?P<0.01 or P<0.05?.3.Compar ed with diltiazem group,the expression of NCX mRNA and myocardial infarction were decreased in the high-and low-dose groups?P<0.01 or P<0.05?,and the calcium i on concentration in the high-dose group was decreased?P<0.01?.4.there was a si gnificant difference between the high-dose group of Xintong Fang and the low-dose group of Xintong Fang.The expression of NCX mRNA,calcium ion co ncentration and myocardial infarction were significantly decreased in the hig h-dose group of Xintong Fang?P<0.01 or P<0.05?.Conclusion 1.The normal expression of NCX in myocardial tissue after myocardial infa rction in rats with acute myocardial infarction may be inhibited,and the mechanism of calcium overload injury is involved.2.It was shown that each drug intervention group can inhibit the expression of NCX mRNA in myocardial tissue of rats with acute myocar dial infarction,inhibit the reverse transport of NCX,participate in the inhibition of calcium overload formation,and alleviate myocardial damage.3.Xintong Fang is su perior to diltiazem in relieving myocardial damage in rats with acute myocardia l infarction,and the high-dose group of Xintong Fang is outstanding.4.Xinton g Fang could down-regulate the expression of NCX mRNA in rats with acu te myocardial infarction,inhibit the damage of myocardial cells induced by calcium ov erload,and reduce the infarct size of myocardial infarction.The high-dose group of X intong Fang is better than the low dose group of Xintong Fang.
