Objective:To investigate the relationship between the therapeutic effect of Xitong Kelifang on KOA and SDF-1/CXCR4 signaling pathway.Methods:Sixty SD rats of 7 weeks old were fed for one week after adaptive feeding.They were divided into 6 groups according to random number table:blank group,model group,glucosamine sulfate group and Xitong Kelifang(low,medium,High concentration)group.The right knee joint of the rats in the blank group was injected with 50ul of normal saline,and the right knee joint of the other groups was injected with 50ul of 40mg/ml sodium iodoacetate solution to establish the KOA model.Confirming the success of the model.Intervention each group with different drugs for 28 days,then take the materials.The right knee joints of each group were stained with HE for morphological observation and the degree of inflammation was evaluated.Serum IL-1?,TNF-?,MMP-3 and MMP-13 were detected by double antibody sandwich enzyme-linked immunosorbent assay(ELISA).The expression of chemokine SDF-1 protein and its receptor CXCR4 in articular cartilage were detected by immunohistochemistry.Results:1.Gross observation and HE staining showed that the articular cartilage surface of the blank group was smooth,the synovial cells were arranged neatly,and no inflammatory cell infiltration was observed.The model group showed obvious manifestations of osteoarthritis.Severe cartilage destruction can be seen under the naked eye.Subchondral bone can be seen in the weight-bearing area.Microscopically,villus epithelial cells proliferate and extend into the joint cavity;cubic epithelial cells and synovial cells proliferate with a large number of vascular proliferation;inflammatory cell aggregation can be seen in the visual field.Fibrosis occurs in the local synovial tissue.Different degrees of cartilage destruction,synovial fibrosis and inflammatory cell infiltration were observed in the other groups,which were lighter than the model group.In the drug intervention group,the degree of joint destruction was the lowest in the group with high concentration of Xitong Kelifang.2.The analysis of the results of ELISA showed that compared with the blank group,the four indexes of the model group and the drug intervention group were obvious.Elevated,P<0.05;and compared with the model group,the four indexes of the drug intervention group were lower than the model group,P<0.05;the positive drug glucosamine sulfate and different concentrations of Xitong Kelifang were found.The mean levels of inflammatory factors and matrix metalloproteinases in group A and group L were similar,P>0.05.The comparison of group A and group M showed that the levels of inflammatory factors and matrix metalloproteinases in group M were significantly lower than those in group A.Except for IL-1? accident,P was less than 0.05;H group was significantly better than group A,P<0.05;comparing the levels of inflammatory factors and matrix metalloproteinases between different concentrations of Xitong Kelifang group,four The indicators had the same trend,namely,L group>M group>H group,M group and L group,P was less than 0.05,and there was no statistically significant difference between M group and H group.3.Analysis of the results of immunohistochemical detection data showed that compared with the blank group,the optical density values of SDF-1 and C XCR4 in the model group and the drug intervention group were significantly higher,that is,the positive cell rate was significantly higher than the blank group,P<0.05;and the drug intervention group compared with the model group,the positive cell rate was significantly lower,P<0.05;the positive drug glucosamine sulfate and different concentrations of knee pain granules were compared and found that the former synovial cells expressed SDF-1 The positive rate and the positive rate of chondrocyte expression of CXCR4 were significantly higher,P<0.05;compared with the cell positive rate of SDF-1 protein and its receptor between different concentrations of knee pain granules,stromal cells were found.The expression of the derived factor-1 and its receptor had the same tendency,that is,the L group>M group>H group,and the H group compared with the L group,P>0.05.Conclusion:Xitong Kelifang can reduce the levels of inflammatory factors IL-1?and TNF-a in OA joints,reduce the cascade of inflammation and inhibit the expression of chemokine SDF-1 in synoviocytes,thus inhibiting SDF-1/CXCR4 signaling pathway,down-regulate the expression of MMP-3,MMP-13,inhibit catabolism,and thus play a role in the treatment of knee osteoarthritis.After comparison between the groups,it was found that the treatment effect of Xitong Kelifang was superior to that of glucosamine sulfate capsules.
