Purpose: Test TRAIL and IL-10 during skeletal muscle apoptosis and try to figure out the related mechanism that TRAIL and IL-10 are involved in the skeletal muscle apoptosis of injured rat and compare the diagrams which elaborate the expression of TRAIL and IL-10 in blood and skeletal muscle during the process of apoptosis.According to the data of observation,try to look for the regulation of change and analyze it with the help of Qi-Blood theory from TCM.With the help of explorative research,detailed mechanism that TRAIL and IL-10 are engaged in the apoptosis during the healing of acute soft contusion without cut as well as the regulation of change by the means of comparing the data from the blood and the skeletal muscle will be disclosed.Creatively applying Qi-Blood theory to the analysis of apoptosis and immune factors is a new insight into the regulation of change during the rehabilitation,which may provide the fundamental evidence to the cut-in time of traumatic treatment and the appliance of Qi-Blood theory.Methods: Randomly select 80 SD rats and adopt the lottery to group the rats with ten rats each,including control group,1 hour group,12 hours group,24 hours group,2days group,3 days group,7 days group and 14 days group.Mold with quantitative strike and kill rats in the control group after all the molding rats.Extract blood and tissues from skeletal muscle as samples.Identify the apoptosis with TUNEL and observe the pathological section.Test the expression of TRAIL and IL-10 in blood and skeletal muscle.After analyzing experimental data,we may explore the regulation of change in the apoptosis and comparatively analyze the curved graphs on blood and skeletal muscle.Finally,combine the Qi-Blood theory with the exploration of the influential elements for those changes.Results: 1)Twenty hours after the injury,the execution of apoptosis is inactivated thoroughly.The expression level of TRAIL in blood is soaring to the peak,while the expression of IL-10 in blood is falling down to the valley.2)Twelve hours to three days after the injury,the apoptosis of wounded tissues is obviously increased and the expressions of TRAIL and IL-10 go parallel,which hints that a pseudo-stable stateappears in pathology.3)Fourteen days after the injury,the level of TRAIL and IL-10 in blood is approaching to the normal,while the level of TRAIL and IL-10 in skeletal muscle and apoptotic rate are not close to the standard.4)The curved diagrams of means of odds ratio for TRAIL and IL-10 have conjugate trend.Conclusion: The curved line of immune factor in blood twelve hours after injury may give us a hint that the balance of Qi-blood in the whole is firstly destroyed.The stable trend of immune factors in blood and skeletal muscle may be administrated by pseudo-stable state of Qi and Blood in whole body and local tissue in pathology,during the execution of apoptosis.Fourteen days after injury,immune reaction is end,however,apoptosis and immune reaction in local are unceasing,which suggests that it is related to the establishment of the new balance of Qi and Blood in whole body and local tissue.The rebalance of Qi and Blood in the whole body and local tissue may have the direct influence on the conjugate trend of immune factors during different periods.
