Blumea balsamifera DC.is a folk herbal medicine,which mainly in the area south of the Yangtze River.Its whole herbs or aerial parts are used as a crude drug for lung cancer in Yi national,because of the characteristics of bitter cold.The main chemical components ofB.balsamifera are volatile oils,flavonoids,terpenoids and sterols,with pharmacological activities such as anti-tumor,anti-oxdation,anti-inflammatory and liver protection.The medicinal meterials was extracted with 90%ethanol,and the ethanol extract was supended with water and fractionated by petroleum ether,methylene chloride,ethyl acetate and n-butanol.Then we used silica gel columnchromatography,LH-20gel column chromatography,rapid preparation or semi-preparative HPLCand other methods to investigate the chemical constituents.Seven compounds were isolated from the whole B.balsemifera,their chemical structures were identified with NMR and MS,and the compounds were indentified as Liquiritigenin(Compound ?);5-hydroxy-7,3',4'-trimethoxydiflavones(Compound?);3,5,7,3'-tetrahydroxy-4'-methoxydiflavones(Compound ?);5,7,4'-trihydroxydihydroflavone(compound ?);kumatakenin(Compound ?);3,5,3'-trihydroxy-7,4'-dimethoxyflavone(compound ?);Diosmetin(compound ?),the compound ? and ? were discovered for the first time in this plant.In the aspect of in vitro anti-tumor evaluation of the compounds,we selected human non-small cell lung cancer A549 cells in this study.CCK-8 method was used to evaluate the in vitro cell proliferation inhibition activity;Scratch method was used to evaluate cell migration activity;PI staining was used to detect the the effect of cell cycle and the effect of ROS on cell apoptosis was evaluated by DCFH-DA staining.The results indicated that 5 compounds had inhibitory activity on the proliferation and metastasis of A549 cells except of compound VI,and the inhibitory effect of compound ? and compound ? at high concentrations was inversely related to the administration concentration.In the evaluation of cell cycle,flow cytometry revealed that compounds ?,?,?,and ? arrested A549 cells in G2/M phase;and the intracellular A549 activity after compound ?,?,?,and IV administration,and there is a significant increase in the concentration of reactive oxygen species in A549 cells which were treated by compounds ?,?,?,and ?.In this paper,the chemical composition of B.balsamiferawas isolated and its structure was identified.The compounds were evaluated for their antitumor activity in vitro and it provides a reference for the further study of their anti-cancer activity.
