Characteristics and clinical significance of RAG1,RAG2 expression in adult B-cell acute lymphoblastic leukemiaObjective:RAG1 and RAG2 played important roles in lymphopoiesis and function of immune system,its dysfunction may led to the occurrence of hemopoietic system malignancies.The aim of this study was to investigate the characteristics of RAG1and RAG2 expression in adult B-cell acute lymphoblastic leukemia(B-ALL)and analyze the clinical significance.Methods:Quantitative PCR(q-PCR)was performed to evaluate the expression of RAG1 and RAG2 in 104 newly diagnosed,22 relapsed adult B-ALL patients and 30normal control,clinical significances of RAG1 and RAG2 expression were analyzed.Mann–Whitney U test and Pearson?~2 test(or Fisher's exact test)were used to compare the clinical and laboratorial parameters between RAG1,RAG2 high expression group and low expression group.We used Kaplan-Meier survival curve to analysis survival differences between RAG1,RAG2 high expression group and low expression group.SPSS software(version 19.0)was used to perform all the statistical analysis.Differences were considered to be significant when P<0.05.Results:Compared with normal control,newly diagnosed and relapsed adult B-ALL patients showed higher RAG1 and RAG2 expression.6 patients with paired newly diagnosed/relapsed samples exhibited higher RAG1 and RAG2 expression when disease relapsed than newly diagnosed.The rate of LDH level above 1000U/L was higher in RAG1 high expression group.Higher RAG1 and RAG2 expression were detected in Ik6 positive group than Ik6 negative group.Patients with high expression of both RAG1 and RAG2 simultaneously showed higher CRLF2 mutation rate than other patients.Conclusion:RAG1 and RAG2 up-regulation may play an important role in the development of adult B-ALL especially when relapsed.It may also take part in the formation of Ik6.Monitor of RAG1 and RAG2 expression may provide a new method for clinical to evaluate disease progress and prospect prognosisPart ?
