Obesity refers to a metabolic syndrome,which abnormal accumulation and distribution of adipose tissue due to abnormal energy metabolism.It is closely related to diabetes,fatty liver,cardiovascular diseases and cancer,and have the high incidence rate and widely the coverage rate,with the treatment has little effect.The phragmites communis(Cav.)Trin.ex Steud is a young bud of the Phragmites communis(Cav.)Trin.blelongs of Gramineae,we call it “Lusun” in chinese,it's rich in phenolic acid,and has potentialits in efficacy of anti-obesity on account of its antioxidant and anti-inflammatory activity,has a unique advantage as a diet supplement.At present,there are few studies on the material basis and mechanism of its edible buds Lusun,which limits the functional development,further application and promotion of the Lusun product.In this paper,we firstly utilized the physical and chemical properties of phenolic acid to prepare phenolic acid extract,determined the content of phenolic acid in the extract,and detected the antioxidant activity of asparagus polyphenol extract.At the same time,the content of some active components in lusun was determined by HPLC,and discusse its material basis.And through the establishment of high-diet-induced obesity mouse model to evaluate asparagus polyphenol extract in vivo experiments on the role of obesity intervention.And through the establishment of high-diet-induced obesity mouse model to evaluate lusun polyphenol extract in vivo experiments on the role of high cholesterol and high blood sugar intervention.In addition,we also tested the anti-preadipocyte differentiation and anti-macrophage activation of lusun phenolic acid by in vitro cell experiments.The experimental results are as follows: 1.The content of phenolic acids in lusun polyphenol extract was determined by UV-visible spectrophotometry,which was 0.532mg/mg.2.The establishment of the determination the contents of 3 phenolic acids,six flavonoids in lusun which was protocatechuic acid,chlorogenic acid,vanillic acid,naringin,rutin,quercetin,luteolin,kaempferol,apigenin by HPLC.3.The DPPH · method was used to determine the antioxidant activity of polyphenol extract.The results showed that polyphenol extract had some antioxidant activity and was close to the positive control vitamin C.4.Through the pharmacological experiments in vivo,the model of obesity in mice was established and the body weight,GLU,TG,LDL,HDL and LDL / HDL were determined.Lusun phenolic acid was found to have the pharmacological activity of controlling body weight and reducing blood glucose and blood lipid.5.In vitro cell experiments found that lusun phenolic acid extract could inhibited the differentiation of 3T3-L1 cells significantly,and dose-proportional.It was found that it can interfere with inflammatory response of activated macrophages induced by LPS,reduce its production of NO,and in a dose-dependent manner.
