| This paper is mainly divided into two parts.The first part is the theoretical analysis.Through the understanding of spleen and its related diseases,brain and brain edema,the paper excavates the correlation of spleen brain in Nei Jin,and puts forward the new idea of "treating encephalopathy from spleen".Exploring the theoretical,scientific and clinical rationality of treating encephalopathy from spleen provides creative ideas and methods for the establishment of syndrome differentiation system of the five Zang organs.At the same time,it also provides the theoretical basis for the clinical application of Jianpi Yiqi Lishui Fang in treating ischemic brain edema.The second part is the experimental research,the specific research is as follows:Objective:To investigate the protective effect of Jianpi Yiqi Lishui Fang on cerebral edema in rats with cerebral ischemia and reperfusion and its effect on the expression of tight junction protein Occludin,Claudin-1 and ZO-1.Methods:168 male SD rats were randomly divided into sham-operated group(n=24)and model group(n=144).The MCAO model was established by the method of thread embolization.After the establishment of the model,the rats were divided into five groups: model group,Edaravone group,high dose group,middle dose group and small dose group.After 7 days,the nerve function defect score was evaluated before execution,the permeability of blood brain barrier was detected by Evans blue evanses blueEBs,the expression of tight junction protein Occludin,Claudin-1 and ZO-1 in brain tissue was detected by immunohistochemistry,the content of Occludin,Claudin-1 and ZO-1 protein in ischemic reperfusion brain tissue was detected by Western blot method,and fluorescence quantitative polymerase chain reaction(RT-PCR)was used to detect the expression of Occludin,Claudin-1 and ZO-1 mRNA in brain tissue after cerebral ischemia-reperfusion.Results:Compared with the sham-operated group,the neural function score increased significantly in the model group(P<0.01),the content of EB in the ischemic brain tissue increased significantly(P<0.01),and the expression of Occludin,Claudin-1 and ZO-1 protein and its m RNA decreased significantly(P<0.01).Compared with the model group,the nerve function score decreased(P<0.05 or P>0.05),and the content of EB decreased in the ischemic brain tissue(P<0.05 or P>0.05),the expression of Occludin,Claudin-1 and ZO-1 protein and its mRNA increased in the positive drug group and Jianpi Yiqi Lishui prescription group.(P<0.01 or P<0.05).Conclusion:Jianpi Yiqi Lishui Fang upregulated the expression of tight junction protein Occludin,Claudin-1 and ZO-1,improved the permeability of BBB,alleviated cerebral edema and improved neural function in rats with cerebral ischemia and reperfusion. |
