Part ?:A Correlative Study of the PON Genetic and Cognitive Impairment after Acute Ischemic StrokeObjective: This study aims to explore the correlation between the single nucleotide polymorphisms(SNP)of the Paraoxonase(PON)gene and cognitive impairment after acute ischemic stroke(CIAAIS).Methods: Using the first acute ischemic stroke as the research object and the nested case as the control study,standard patient information,including personal details,medical history,blood biochemical indicators,was collected and used for evaluation of patients cognitive function after acute ischemic stroke(2 weeks(14 days)after disease onset)by neuropsychological assessment.Then,the nine polymorphic locus genotypes of PON gene were measured and analyzed.Moreover,the incidence of cognitive dysfunction after acute ischemic stroke,and the correlation of PON gene polymorphisms genotypes and CIAAIS were evaluated through the statistical correlation analysis.Meanwhile,subgroup analysis was carried out according to the gender and the degree of cognitive impairment.The influence of the interaction between PON gene and environmental risk factors on the risk of CIAAIS was also evaluated.Results:(1)The incidence of cognitive impairment was 49.01% after the onset of acute ischemic stroke for 2 weeks.Atrial fibrillation(AF)was significantly associated with CIAAIS(P<0.05).(2)CC genotype of rs7493 were significantly associated with CIAAIS in total population and mild cognitive impairment subgroup,and significantly increased the CIAAIS susceptibility.(3)The influence of the interaction between PON gene and environmental risk factors on CIAAIS occurrence was not significantly observed in the analysis of total population and subgroup.Conclusions:(1)Atrial fibrillation is a risk factor for CIAAIS.(2)SNP in PON genes is associated with CIAAIS.Also,the risk of cognitive impairment increase after cute ischemic stroke for individuals carried rs7493 CC genotype.Part ?:A comprehensive systematic review with Meta?analysis on the relationship of the PON gene and Alzheimer's diseaseAim: This study aimed to evaluate the association between the PON genes variants and AD using systematic review with meta?analysis;Methods: Relevant studies were identified by searching English and Chinese databases extensively.Allele and genotype frequencies for each included study were extracted.Newcastle?Ottawa Scale(NOS)was employed to evaluate the quality of included studies.The odds ratio(OR)was calculated using a random?effects or fixed?effects model.A Q statistic was used to evaluate homogeneity,and Egger's test and funnel plot were used to assess publication bias;Results: A total of 15 studies were included and identified for the current meta?analysis.The NOS scores ranged from 7 to 8,meaning good quality of studies.It was found that the SS genotype of PON2 S311 C polymorphism had an significant association with AD in the studied population(OR=0.82,P=0.04),However,there was no significant relationship between other three genetic variants of PON genes and AD;Conclusions: Existing evidence indicated that the S311 C polymorphism(SS genotype)was associated with risk of AD in studied population.Future studies with larger sample sizes will be necessary to confirm the present results.
