Objective:to establish the successful animal model of subarachnoid hemorrhage as a method to observe the expression of T type calcium channel ?1G in cerebral vascular model in rats at different levels,and through specific antagonist intervention,Objective to investigate the significance of T type calcium channel in the pathogenesis of delayed vasospasm after subarachnoid hemorrhage,and to further explore the mechanism of injury and pathological basis,so as to provide a new method for clinical treatment.Methods:Through the chiasm blood injection,the establishment of a rat model of subarachnoid hemorrhage,combined with Garcia JH neurobehavioral score and HE staining effectiveness evaluation of rat nerve function damage degree and model.Western blotting,Quantitative Real-time,immune at different time to observe the expression in rat model of ?1G in cerebral vessels in use,and the application of specific calcium channel antagonist nimodipine and flunarizine hydrochloride intervention,neurobehavioral score,using Garcia JH HE staining and Western blot assay the expression of different time in the nerve function damage degree,the degree of vascular spasm,?1G judging the therapeutic effect evaluation.Results:1.The overall mortality rate was 30%,and the postoperative neurological score was lower than that of saline injection group and normal group.The Garcia J H score after operation showed that there was a decrease in activity and a dec rease in response.The intervention group rats was stronger than chiasmal bloo d injection group,the behavior of flunarizine hydrochloride group was higher t han the score of the rat nimodipine group,early nimodipine group rats was hig her than the score of flunarizine hydrochloride group.2.According to the model of cross injection of blood,the blood can be distributed on the surface of the brain.The brain tissue sections of the rat model of cross injection showed that the cerebral vessels in different periods had different degrees of spasticity,and the degree of vasospasm of the first 2day was the most serious.The intervention group is less than the degree of vascular spasm chiasmal blood injection group,nimodipine group 2day vasospasm is less than corresponding days of flunarizine hydrochloride group,the 6day group and flunarizine hydrochloride vascular spasm degree is smaller than those of the corresponding number of nimodipine group,Ca ion channel antagonists for vasospasm after subarachnoid hemorrhage treatment,T type calcium channel antagonist flunarizine hydrochloride group and the remission of delayed vasospasm was better than that of L type calcium channel antagonist nimodipine,and nimodipine for early termination of vasospasm is better than flunarizine hydrochloride group.3.Protein electrophoresis observed ?1G is abundantly expressed in cerebral vessels of rats in the model,the number is far higher than that of normal rats,especially in the accumulation of blood,such as the most concentrated expression of the basilar artery.There was no significant difference between the intervention group and the control group.4.real time fluorescence immunoassay PCR showed that the expression of Cav3.1mRNA in normal group,saline group,blood injection group,the intervention group had the blood injection group,the content of Cav3.1mRNA in the intervention group were higher than that in normal group,no significant difference between the intervention group and blood injection group.5.All the blood injection model rats had weight loss,with no postoperative weight of intervention group rats decreased significantly,the ability of anti early nimodipine decreased body weight than flunarizine hydrochloride,and flunarizine hydrochloride against advanced weight loss is greater than the capacity of nimodipine.Conclusions:1.In rats with subarachnoid hemorrhage,the expression of T type calcium channel alpha 1G subunit in cerebral blood vessels increased progressively in 6 day.2,Nimodipine and flunarizine hydrochloride is not through the change of T type calcium channel expression play a therapeutic effect.
