Pancreas development process is a complex and conservative process with a series of signaling pathways and transcription factors.etsl and ets2 belongs to the ets transcription factors,which can promote cancer cells migration and invasion,blood vessel formation and differentiation of lymphocytes,participate in organ development.In recent years,ets1 and ets2 are also found to be available to play a role in the pancreas cells,such as the etsl not only play a role in the normal pancreas cells,but also played a part in the process of pancreatic cancer,and in mice pancreas development,ets2 can strengthen cell proliferation,differentiation of the pancreatic acini and ducts.But we know little about the role of both ets1 and ets2 in the process of embryonic development.This experiment uses zebrafish as a model to study the function of ets1 and ets2 in the process of pancreas development.It was found that single knock down etsl or ets2 has no effect on zebrafish embryonic development,but the common knock down etsl and ets2 causes the deformity of zebrafish embryos.In zebrafish pancreas development,ets1 and ets2 absence has no effect on the expression of pdx1 during the early development of pancreas;in pancreatic endocrine development period,the expression of pancreatic endocrine marker ins quantity is increased;in the exocrine pancreas development period,the pancreas exocrine cells marker try expressing quantity increases.But the lack of both does not affect the the expression of liver,small intestine and stomach markers.Bioinformatics analysis and the double luciferase reporter gene experiment found etsl and ets2 can combine to igfl promoter region.Igf1 can promote cell mitosis,inhibiting cell apoptosis function in different types of cells at the same time.Our experiments prove that knocking down the etsl and ets2 raised expression of igfl,zebrafish embryos after overexpressing igfl appear the similar pancreas phenotypes to the knocking down of ets 1 and ets2 group.knocking down igfl can relieve the pancreatic abnormalities of the lack of etsl and ets2.All results above prove that etsl and ets2 may combine to igfl promoter region to regulate the development of pancreas in zebrafish embryos in the early stage of the zebrafish endocrine cell development,and play a role in the process of differentiation during pancreatic development.
