Objective:(1)To evaluate the influence of anti-drug combination on material transport and barrier function of vitro intestinal tissue and provide experimental evidence for the theory of incompatibility.(2)To evaluate the influence of anti-drug combination on the absorbing,secretion and barrier function of gastrointestinal tract of spleen-deficiency rat model and provide experimental evidence for the theory of incompatibility.Method:(1)Ussing-chamber technique was adopted to evaluate the functions of vitro intestinal tissue,among which FD4 and R123 were used to evaluate its material transport function,and FD40 was used to evaluate its barrier function.(2)Reserpine intraperitoneal injection was used to build spleen-deficiency rat model.The body weight,food intake and mental state of these rats were scored,and SOD and MDA level of gastric tissue were applied to evaluate their spleen deficiency symptoms.The absorption function of rats was evaluated by D-xylose uptake and Na+-K+-ATP activity,and the secretion function of the gastrointestinal tract was evaluated according to the gastrin,motilin,vasoactive intestinal peptide and somatostatin level in serum.Diamine oxidase and endotoxin level in serum and average barrier function of rats were determined.Results:(1)Licorice could significantly promote the absorption and secretion of R123 FD4,and they were significantly suppressed after combining licorice and"sargassum,euphorbia pekinensis,kansui,and genkwa,.Licorice had no significant effect on the penetration of FD40,and it increased after combining algae,kansui and licorice.When euphorbia and daphne were separately used,the penetration of FD40 significantly increased compared with the control group,and it significantly decreased after licorice compatibility.(2)It can be found from the overall state scoring of all groups of rats that reserpine modeling could lead to a significant increase of spleen index,and the spleen index of licorice group was significantly lower than the model group.The monotherapy group of "sargassum,euphorbia pekinensis,kansui,and genkwa,had no significant difference compared with the blank group;the spleen index of combination group was greatly improved,but the symptoms were severe eombined with the licorice group;the spleen indexes of the euphorbia and licorice combination group as well as the daphne and licorice combination group were significantly higher than the licorice group.SOD level of model group rats significantly decreased combined with the blank group,and MDA level significantly increased.Apart from euphorbia group,SOD level of other medical treatment groups signifieantly increased combined with model group.Licorice could decrease MDA level of stomach tissue of spleen deficiency rats.Algae,euphorbia and kansui had no significant effect on the MDA level of stomach tissue of spleen deficiency rats,and after the combination of licorice,algae,euphorbia and kansui,the MDA level of stomach tissue of spleen deficieney rats had different degrees of deeline,in which the MDA level of the algae and licorice combination group significantly decreased.(3)D-xylose excretion of spleen deficiency rats significantly decreased compared with the blank group,and after licorice intervention,it significant increased compared with the model group.On the condition of separately using algae,euphorbia and kansui and daphne alone,they had no significant influence on the D-xylose excretion of spleen deficiency rats.When combining algae,euphorbia,kansui and daphne with licorice,the D-xylose excretion comparatively increased compared with the monotherapy group,and it significant increased after the combination of euphorbia and licorice compared with the model group.Na+-K+-ATPase activity of the model group significantly decreased compared with the blank group.Licorice could significantly improve Na+-K+-ATPase activity of jejunum issue of spleen deficiency rats.Algae,euphorbia,kansui and daphne could also significantly improve the Na+-K+-ATPase activity.After licorice was combined with "sargassum,euphorbia pekinensis,kansui,and genkwa",the combination of algae and licorice could significantly improve the Na+-K+-ATPase activity,and it had an upward trend compared with the licorice group.After combining euphorbia,kansui and daphne with licorice,Na+-K+-ATPase activity significantly decreased compared with licorice group although it significantly increased compared with the model group.(4)MTL,GAS and VIP level of rats in model group significantly decreased compared with the blank group,and the SP level significantly increased.Licorice could reverse the MTL,GAS and VIP secretion down regulation and SP secretion up regulation.Algae,euphorbia,kansui and daphne could also improve the MTL and GAS level to varying degrees.After licorice was combined with"sargassum,euphorbia pekinensis,kansui,and genkwa",MTL levels all dramatically increased in addition to the licorice and daphne combination group.GAS level in the combination groups had a trend of increasing,among which the GAS levels of euphorbia and licorice combination group as well as daphne and licorice combination group decreased significantly combined with the licorice group.Algae,euphorbia and daphne had no significant effect on the VIP level,and after licorice was combined with "sargassum,euphorbia pekinensis,kansui,and genkwa,,VIP levels of other eombination groups all dramatically increased in addition to the algae and licorice combination group.Algae,kansui and daphne could also reduce the SP secretion of spleen deficiency rats.After the combination,SP levels of all the combination groups comparatively increased combined with the licorice group,and the euphorbia and licorice combination group had significant difference with licorice group.(5)Compared with the blank group,the endotoxin of serum in the model group rats significantly increased.Under licorice intervention,the endotoxin of serum significantly decreased.Algae,euphorbia,kansui and daphne could also reduce the endotoxin level of serum in spleen deficiency rats.When combining licorice with"sargassum,euphorbia pekinensis,kansui,and genkwa",although endotoxin levels of serum in all groups of rats decreased signifieantly eompared with model group,the endotoxin level still significantly increased combined with the single group of licorice.Under licorice intervention,DAO level of serum in spleen deficiency rats significantly decreased.Algae,euphorbia,and kansui could also greatly reduce the DAO level.When eombining licoriee with“sargassum,euphorbia pekinensis,kansui,and genkwa",DAO levels of all the combination groups increased to varying degrees compared with the licorice group,among which the DAO levels of euphorbia and licorice combination group as well as and daphne and licorice combination group dramatically increased compared with the licorice group.Conclusion:(1)Licorice significantly promotes the material transport capacity of intestinal tissue in normal rats,and has no significant effect on the barrier function of the intestinal tissue,which is probable because licorice has functions of drug ingredients absorption and detoxification.When combining licorice with"sargassun,euphorbia pekinensis,kansui,and genkwa",the promoting function of licorice was significantly suppressed,and the barrier function of intestinal tissue was also damaged to varying degrees,indicating that after the combination,pharmacological effects of licorice received antagonism and that there emerged the incompatibility and efficiency reduction of licorice after the co-using of "sargassum,euphorbia pekinensis,kansui,and genkwa".(2)According to the comprehensive evaluation of the symptom and gastrointestinal function of spleen deficiency model rats,licorice can significantly improve their symptom and gastrointestinal function,which reflecting its effect of"replinishing qi to invigorate the spleen,.After the combination,the efficacy of anti-drug combination on their symptom and gastrointestinal function is lower than licorice,which shows that pharmacological effects of licorice received antagonism and reflects the incompatibility and efficiency reduction of licorice.
