SPinach(Spinacia oleracea L.)is a rich variety of vitamins and minerals in vegetables,but vulnerable to a variety of pathogens,resulting in lower yields,the quality of economic decline and other issues.Among them,Spinach downy mildew is a major disease problems affecting the production Spinach cultivation.Spinach downy mildew is one kind of fungal diseases caused by obligate pathogens(PeronoSPora farinosa f.Sp.Spinaciae,Pfs).The main hazards Spinach leaves,Spinach occur in the seedling stage,flowering stage and the adult stage can be widely popular in the world,seriously affecting production Spinach.Currently Spinach downy mildew prevention mainly through agriculture and prevent chemical control,chemical control,although effective concentration,but because of drug-resistant pathogens and food safety issues,it continues to explore safe and effective means of preventing.Probenazole(PBZ)is an effective control of rice blast on a novel anti-chemical attractant which acquired resistance by salicylic acid(SA)route guidance system.PBZ applied to the study of this study will improve the Spinach downy mildew resistance,downy mildew induced by determining PBZ susceptible Spinach,further screening of the PBZ induced susceptible resistant varieties,explore the PBZ inducing Spinach downy mildew resistance the molecular mechanisms.Great theoretical and practical significance for the cultivation of excellent resistant varieties of Spinach,to build a new chemical induction system.The study was a large-scale field disease resistance germplasm resources survey,completed the downy mildew resistance analysis of germplasm material screened out 14 copies downy mildew resistance significantly different material.Screening of resistant and susceptible materials were PBZ processing measured physiological parameters of Spinach PBZ material after processing includes POD activity,MDA content and soluble proteins,and nutritional indicators such as Spinach oxalic acid,nitrate,vitamin C,physiology and nutrition from two angles to assess the impact of PBZ nutritional quality of Spinach and so on.The results show that,as PBZ no significant impact on the nutritional quality of Spinach,but PBZ growth has some impact on the development Spinach,and Spinach may be related genes and the role of the body.14 different anti selected,susceptible material,for 8% PBZ drench treatment,downy mildew(Race 9)inoculation experiment,observed PBZ process can induce a certain degree of resistance,but different materials induced PBZ there are differences in susceptibility,susceptible material PBZ after induction of disease resistance to improve significantly affected.The observation that high concentrations of PBZ treatment did not accelerate the aging process Spinach.To further explore the molecular mechanism of PBZ induced resistance to downy mildew of Spinach,the present study analyzed the expression pattern in the course of the Spinach treated PBZ genes by realtime PCR data and bioinformatics collaborative filtering PBZ induced reSponse,the results It shows,PBZ significantly induced the expression of genes NPR1 Spinach.Match found NPR1 of bZIP family,the present study analyzed the 58 family members in Spinach genome-wide identified through bioinformatics analysis system development,and from the Spects of cis-acting elements for the conserved domain and other aSPects prediction analysis of the bZIP family members functions,screening candidate genes related to SPinach downy mildew resistance.This study is to explore the molecular mechanism for PBZ and PBZ Spinach downy mildew induced resistance induced preliminary analysis,and screened for using PBZ induced resistance material,in order to further explore new chemical inducers and PBZ Spinach resistance reaction synergies that increase efficiency and disease resistance means resistance breeding Spinach provides a new idea,and laid the theoretical foundation for the Spinach resistant molecular mechanism and biological functions of key genes.
