Optical microscope,as an indispensable experimental tool for life science research,a major challenge in its technological evolution is how to resolve the irreconcilable contradiction between imaging resolution and field of view.The field of view observed under a low magnification objective is large,but the image resolution is insufficient.Under the high magnification objective lens,the image resolution is high,but the observation field of view is limited,and the observer cannot see a wide range of fields of view and deep depth of field images.In this context,the paper has done the following research:Firstly,research on automatic stitching algorithm for calculating microscopic image.In the current image registration and image fusion algorithms,the selection of a suitable algorithm enables the calculation of faster speed while ensuring the accuracy of registration,and finally enables real-time image automatic stitching.Secondly,algorithm for calculating the depth of field development of microscopic imaging.The main research is to obtain a large depth of field image and realize the depth of field expansion when observing static cells without losing the lateral resolution.After that,autofocus algorithm research.Automatic focusing of images is achieved by means of software-driven hardware platforms.A suitable focus evaluation function and an autofocus search method are selected,and an improved scheme is proposed.Finally,software system design.Through detailed analysis of the requirements of the software system,the interface design of the software,the realization of the functional modules,and finally the functions of autofocus,image stitching and depth of field expansion are realized.Based on the analysis of the image splicing and depth of field expansion of the microscopic imaging system,combined with the system design and experiment,through the further improvement of the algorithm,the automatic image acquisition,image automatic splicing and depth of field are realized by software-driven hardware.The function of the extension has effectively broadened the imaging field of view and the depth of field,further driving the practical and industrialization of the technology.
