Because of its powerful analytical capabilities,High performance liquid chromatography(HPLC)has been one of the most important applications in modern methods for separation and analysis of complex mixtures.It is now widely used in analytical chemistry,clinical medicine,environmental monitoring,food safety and other fields.However,due to the complexity and low content of the components to be tested,and the lack of a suitable universal detector,the application of high performance liquid chromatography is also limited.Chemical derivatization can greatly improve the sensitivity of detection and separation conditions,enabling high performance liquid chromatography to detect traces of analytes in the environment.Therefore,research and development of new derivatization reagents have made it a hot research topic in high performance liquid chromatography systems.The focus of this paper is to study the synthesis and application of three new fluorescent-derived reagents.In addition to the improvement of chromatographic conditions,high performance liquid chromatography for the detection of various actual samples has been established.The specific work is as follows:Chapter one: The application of fluorescence derivatization in high performance liquid chromatography-mass spectrometry,including high performance liquid chromatography-mass spectrometry,development and basic knowledge of fluorescence derivatization,is briefly introduced.Finally,the application of thiol,phenol,and amine fluorescent derivative reagents is briefly introduced.Chapter two: Three novel fluorescent-derived reagents: 1-(4-(1H-phenanthro-[9,10-d]imidazol-2-yl)phenyl)-1H-pyrrole-2,5-dione,2-((4)-(1H-phenanthryl[9,10-d]imidazol-2-yl)phenyl)(methyl)amino)ethyl-1H-imidazole-1-carboxylate,4-(1H-p henanthrene[9,10-d]imidazol-2-yl)benzaldehyde synthesis and design,and their respective spectral properties and structures were characterized.Chapter three: 1-(4-(1H-phenanthro[9,10-d]imidazol-2-yl)phenyl)-1H-pyrrole-2,5-dione as a fluorescent derivatization reagent combined with high performance liquid chromatography-mass spectrometry The mercaptan in the food was measured.The mercaptan is enriched by a gas stream purge-injector microextraction system and the purification and derivatization conditions and chromatographic separation conditions are optimized.The detection effect fully satisfies the re-quirements for the establishment of thiol detection method and is applied to t e detection of thiol content in four foods.Chapter four: 2-((4-(1H-phenanthryl[9,10-d]imidazol-2-yl)phenyl)(methyl)ami no)ethyl-1H-imidazol-1-carboxylate as a fluorescent derivative reagent The ultraviolet absorber in the wastewater was determined by high performance liquid chromatography-mass spectrometry.The analytes were purified by liquid-liquid dispersion microextraction,and the purification and derivatization conditions were optimized.A high performance liquid chromatography method for the detection of UV absorbers in wastewater was established.Chapter five: 4-(1H-phenanthro[9,10-d]imidazol-2-yl)benzaldehyde was used as a fluorescent derivatization reagent combined with high performance liquid chromatography-mass spectrometry to analyze and identify aniline.The derivatization conditions and separation conditions were optimized,and the method was applied to the detection of aniline in wastewater.
