Cancer is one of the major disease harm to people’s life and health,while excision surgery,radiotherapy and chemotherapy could not be neglected the risk of side effects.With the rapid development of nanotechnology,the new nanotechnology tumor based on targeting therapy has become a hot research field of cancer treatment.Nano technology and application of RNA interference technology has become a new way for which gene and drug delivery into cells and showed a certain potential in the field of multi-drug resistant.Compared with traditional chemotherapy,co-delivery genes and chemotherapy drugs by increasing the number of drugs to tumor treatment of multiple effect together would obtain good therapeutic effect.In addition,along with the development of research on micro RNA,more and more people know about that micro RNA with great potential in cancer treatment.The small interfering RNA(si RNA)nucleic acid drugs,in the treatment of cancer,has brought new possibilities.Select the appropriate genes(Immediate early genes,IE)will focus on the gene of si RNA into tumor cells,to lower its expression which could safely and effectively inhibit the growth and metastasis of tumor.What’s more,it is because that not all seemingly "perfect" target genes can make si RNA silence directly,when the directly interfere with the expression obstacle,gene could bypass the first purpose,for the second purpose synthetic gene(genes)death for interference suppression,on the premise of no injury to normal cells,to achieve therapeutic effect,thus reduce the side effects of cancer treatment.micro RNA in this paper were introduced from the reported journal articles which with the great ability to kill and inhibit metastasis of three negative breast cancer at the genetic level.Including the following gene CXCR4,micro RNA-205,micro RNA-221,small interfering RNA(si RNA)si RNA-182,si RNA-205 introduced in the papper.To transfer genes to the treatment recognition site,we designed typicle genes to form a single straight DNA and through rolling circle transcription(RCT)to produce treatment gene on a large scale,so as to achieve fast and at low cost but get a lot of nanoscale drug gene system and kill tumor cells effectively.DNA/RNA in the circulating process was easy to degradation and failure to strengthen its circulation and compression.In the meantime,from the RCT we got large size inevitably,in the design of linear DNA transcription template,we design on the cholesterol at the 5 ’end to realize the full geneticsupport hydrogel carry therapeutic genes into cells and mice diagnosis and integration of the tumor cells.In addition,with the deepening of the research,hydrogel as a carrier of the infiltration of tumor cells,the chemical medicines DOX,photosensitizer Ce6 and used to produce oxygen improve tumor microenvironment of manganese dioxide nanoparticles were introduced in the implementation of three negative multi-effect combination therapy of breast cancer.Experimental results show that the targeted nanoparticle carrier mediated has three negative breast cancer gene to targeted cells,inhibit cancer cell growth and metastasis by inhibiting gene expression and promote its apoptosis has more obvious effect.This study based on nanotechnology for negative breast cancer treatment technology has carried on the beneficial exploration. |
