| In the first chapter,the structure types and biological activities of compoun-ds from Peicillium were summarized.The second chapter introduced the ideaa-bout experiment design.In this chapter,one strain Penicillium MCT-21 was sel-ected from the 35 strains from Rhizoma Paridis Yunnanensis by chemical scre-ening with TLC methods.The third chapter mainly introduced the secondary metabolites isolated from Penicillium MCT-21,and the bioactivities of compounds had also been tested.9 compounds were obtained from the endophyte of Polyphylla MCT-21 by a varie-ty of modern separation technology.On the basis of spectral data including of M S,1D NMR and 2D NMR,their structures were elucidated:terrestrol I(1*),(E)-a scladiol(2),(Z)-ascladiol(3),gentisyl alcohol(4),3-hydroxybenzyl alcoho(5),(22E,24R)-ergosta-5a,6a-epoxide-8,22-diene-3p,7a-diol(6),5a,8a-epidioxy-ergosta-6,22-dien-3β-ol(7),7-hydroxy-4,6-dimethylphthalide(8),succinic acid(9).Compound 1 was a new compound.Antibacterial activity test showed that compound 4 exhibi-ted obvious antifungal activity against Bacillus subtilis with MICs at 32 μg/mL.Compounds 1 and 4 exhibited obvious antifungal activity against Staphylococcus aureus with MICs at 64 μg/mL,64 μg/mL,respectively.Compounds 4 exhibited obvious antifungal activity against Escherichia coli with MICs at 128 μg/mL.The fourth chapter mainly introduced the chemical screening of Polyphyllin metabolized by Penicillium MCT-21 by HPLC.The Penicillium MCT-21 was fermented for 5L culture.The mycelia was separated from the supernatant.The mycelia extracted by acetone was concentrated as YYb-1.The mycelia extracted by methanol was concentrated as YYb-2.The supernatant extracted by methanol was concentrated as YYb-3.HPLC was used to test Polyphyllin exit or not in YYb-1,YYb-2 andYYb-3 from Penicillium MCT-21.The fifth chapter mainly introduced the production of metabolites from Metar thizium sp.MCT-12 induced by the steroidal saponin of its host plant,Paispoly phylla.Four compounds were isolated and determined as 4-(2-hydroxyethyl)1,3-benzenediol(1),pennogenin-3-a-L-rhamnopyranosyl-(1→2)[a-L-rhamnopyranosyl(1→4)-β-D-glucopyranoside(2),(4S)-4,8-dihydroxy-3,4-dihydro-1(2H)-naphthalenon-e-l-one(3),polyphyllin VI(4).Compound 1 was isolated from Metarrhizium sp.MCT-12 at the first time.Antibacterial activity test results showed that compoun-ds 1,2 and 3 exhibited moderate antifungal activity against pathogenic bacteria e-xcept for Staphylococcus aureus.The mixture compounds of 1,2,3 exhibited mod-erate antifungal activity of synergistic effect against Staphyloco-ccus aureus with MICs at 128 μg/mL,other pathogenic bacteria antibacterial act-ivity is not obvio-us.The mixture compounds of 1,2exhibited no antifungal activi-ty of synergistic effect. |
