Environmental microorganisms are one kind of important microbial resources,which are the main promoters in the biogeochemical cycle and play the critical roles in construction and maintenance of the sustainable ecosystem.Detection and collection of the microbial gene information from various environments using metagenome-sequencing method and identification of the bacterial cultures with ecological function from some specific environments based on their enzymatic characteristics will provide us with novel thoughts in exploration of microbial resources,which is of significant research prospects.Based on the previous studies in our lab,this study mainly focused on detection of endoglucanase genes and alkaline phosphatase genes,screening of endoglucanase producing strains and phosphorus soluble strains,and carrying out taxonomic studies on the functional strains.Accordingly,to collect the functional genes' information and the gifted bacteria resources.1)Using metagenome-sequencing method,a total of 220 endoglucanase genes belonging to 23 endoglucanase-gene types were detected from the environments where were rich of cellulose.Among which,GH5 endoglucanase genes and GH9 endoglucanase genes were the main components,with the abdunance of 1.14E-02%and 4.6 E-03%,respectively.The taxonomy information of the bacterial resources that contained these genes were acquired by retrieving in the databases,showing bacteria with GH5 endoglucanase gene and GH9 endoglucanase gene affiliation to 6 phyla-12 classes-17oders-30 genera and 6 phyla-10 classes-10 oders-9 genera,respectively.Using CMC-Na as the sole carbon source medium,in total of 113 endoglucanase producing bacterial strains were screened from 410 newly isolated bacteria.These strains with endoglucanase activity affiliated to 3 phyla-13 oders-28 genera.Among these endoglucanase positive strains,strain CPCC 204705T showed 98.54%similarity of 16S rRNA gene with its closest phylogentic neighbor--Cellulomonas oligotrophica JCM 17534T.In the genome sequence of strain CPCC 204705T GH9,GH6 and GH5 endoglucanase genes were found.2)A total of 69 alkaline phosphatase genes were detected,belonging to alkaline phosphatase D gene(phoD)and alkaline phosphatase A gene(phoA),with the abundance of 3.58E-03%and 2.58E-03%,respectively.The bacteria with alkaline phoD and phoA affiliated to 7 phyla-10 classes-14 oders-15 genera and 4 phyla-8 classes-10 oders-8 genera,respectively.With the primary screen method of using egg-yolk as the only phosphorus resource and re-screen method of using orgmiophosphorus-broth,53 phosphorus-soluble strains were confirmed from 583 bacteria.These phosphorus-soluble bacteria affiliated to 3 phyla-4 classes-8 oders-15 genera.Strain CPCC 204705T was detected with alkaline phosphatase activity and contained phoA genes in its whole genome.3)The gifted strain CPCC 204705T,with both endoglucanase and alkaline phosphatase activities,was systematically studied.The polyphasic taxonomy results revealed that strain CPCC 2047051 represented a novel species of the genus Cellulomonas.Since the strain was isolated from a soil sample,we named it as Cellulomonas telluris nov.sp.,according.
