Schizosaccharomyces pombe mitochondrial proteins Ppr10 and Mpa1 are very important for the synthesis of mitochondrial respiratory chain proteins.Ppr10 forms a complex with Mpa1,but how interacts Ppr10 with Mpa1 is not clear.The bioinformatics analysis software HHpred and HHrepID predicting that Ppr10 has13 repeats,these repeats consist of 35 amino acids and are rich in leucine.Due to the similarity between Ppr10 and PPR proteins and between Ppr10 repeats and PPR motifs,so they are named PPR-like motif.To investigate the importance of these PPR-like motifs for Ppr10 function,mutant strains that lacking single or multiple PPR-like motifs are constructed.The interaction of these Ppr10 mutants with Mpa1 and the function of the Ppr10 mutants are analyzed.Research findings in this paper,C-terminal truncation results show that Ppr10 truncated to the No.9 PPR-like motif does not affect mutant strains growth on glycerol medium,the Ppr10 mutants expression are normal and dose not affect the interaction of the Ppr10 mutants with Mpa1.Ppr10 truncated to the No.8 PPR-like motif seriously affect mutant strains growth on glycerol medium,the Ppr10 mutants expression are not normal and destroy the interaction of the Ppr10 with Mpa1.Knock out a single PPR-like motifs results show that the absence of the 1-4 PPR motifs respectively does not affect the interaction between Ppr10 and Mpa1,but affects the growth of mutant strains.The absence of the 5-8 PPR motifs respectively destroys the interaction between Ppr10 and Mpa1 and affects the growth of mutant strains.The absence of the 9-12 PPR motifs respectively affects the interaction between Ppr10 and Mpa1,but does not affects the growth of mutant strains.This study shows,the N-terminal encoded protein of 320 amino acids of Ppr10 is normal expression and is important for the normal function of Ppr10.Among the 13PPR-like motif of Ppr10,the 1-8 PPR-like motif are critical for Ppr10 to functions normally.The 5-8 repeats are essential for the interaction of Ppr10 with Mpa1.The 9-12PPR-like motif are important for the binding of Ppr10 and Mpa1.This paper lays the foundation for further research on the structure and function of Ppr10.
