Traditional methods to prepare biochips for studying interaction between proteins and biological macromolecules need to sample separatation and purify which is time-consuming and completely changes the original biological environment of sugar and protein.Thus traditional methods can not provide fully and true information of the interaction.While cell sensor technology based on quartz crystal microbalance(QCM)can detect cells in real-time and label-free,and obtain real information of interaction between biological molecules at cellular level.Currently,the cells sensor preparation method exists many problems such as weak fixed strength,survival rate,and easy shedding.To solve the above problems,this paper studied two strategies for preparing suspension cells based on QCM technology.1)We modified the cell surface with the clickable group by the cell metabolism for non-natural sugars probe,and prepared the suspension cell sensor by immobilizing the cells on sensor sensor surface through click reaction.2)We coated the QCM gold sensor with polydopamine through the oxidation polymerization of dopamine,and then we prepared the suspension cell sensor by immobilizing the cells on sensor sensor surface through Michael addition reaction between amine and thiol groups.By comparing the above-mentioned two different suspension cell sensor methods through observing the immobilized condition and the cell viability in the continuous phase,we finally established the QCM suspension cell sensor preparation process base on covalently immobilized cells Michael addition reaction We prepared Jurkat and K562 suspension cell sensor by using this method and studied the interaction of cell surface glycosylation and wheat germ agglutinin(WGA),concanavalin A(Con A),peanut agglutinin(PNA),Ulex europaeus agglutinin-I(UEA-I)and soybean agglutinin(SBA),and preliminary studied the tumor cell surface glycosylation.This research provides a fast,efficient,convenient and simple way to the study of glycosylation on cell surface in situ,which is important for the study of biological macromolecules interaction based on QCM technology.
