1 ObjectiveLong-term high fat diet increases the risk of cardiovascular disease,this study uses UAS-Gal4 system to express Sod2 genes by exercising in high-fat diet drosophila,detects TAG,dfoxo,Sod2 gene expression and heart function-related indicators.This paper expounds the effect ofexercise on the heart function of high-fat diet drosophila,and discusses the mechanism of exercise to improve the heart function of high-fat diet drosophila,so as to provide theoretical basis for the study of exercise and its pathways.2 MethodsSod2 overexpression drosophila was obtained by UAS-GAL4 system,and it was cross-breeded with FoxO?94 to obtain foxo missing rescue group drrosophila.Collect 1200 of the three virgin drosophila of FoxO?94?4-?,FoxO?94/UAS-Sod>arm-Gal4?4-2-A?,and W1118>arm-Gal4?W-A?.Each was randomly divided into four groups:the Normal Diet Control Group?NF-C?,the Normal Diet Exercise Group ?NF-E?,the High Fat Diet Control Group?HFD-C?,and the High Fat Diet Exercise Group?HFD-E?,for a total of 12 groups.Each high fat diet group was high fat at the second day of age.The sports team all performed 1.5h exercise intervention on the 16th day of age,lasting two weeks,five days of exercise a week and two days off.All groups were tested for TAG content at a 30-day age to detect dfoxo and Sod2 gene testing,and heart function.3 Results3.1 TAG Level Detection resultsIn W-A drosophila,the HFD-C group TAG/weight?P<0.01?was significant compared to the NF-C group.The HFD-E geoup TAG/weight?P<0.01?was significantly lower than the HFD-C group.In 4-drosophila,the NF-C group TAG/weight?P<0.01?was significantly higher than in the NF-C group in W-A.In 4-drosophila,the TAG/weight level in the HFD-C group was significantly higher than that of the NF-C group?P<0.01?.In 4-2-A drosophila,the HFD-C group TAG/weight?P<0.01? was significantly higher than in the NF-C group.The HFD-E group TAG/weight?P<0.01?was significantly lower than the HFD-C group.3.2 Heart function indicator test resultsIn W-A drosophila,HFD-C group fibrous fibrillation?P<0.05?was significantly higher,and the shortened fraction?P<0.05?and diastolic diameter?P<0.01?were significantly lower than those of NF-C group.The professed fibrillation?P<0.01?in HFD-E group was significantly lower,and the shortened fraction?P<0.01?and diastolic diameter ?P<0.01?were significantly higher than those of HFD-C group.4-Drosophila in the NF-C group diastolic diameter?P<0.05?,shrink diameter?P<0.05?is significantly low,fibrous fibrillation?P<0.05?is significantly higher than the NF-C group in W-A.In 4-drosophila,the HFD-C group fibrous fibrillation?P<0.05?was significantly higher than that of the NF-C group,and the fibrous fibrillation in HFD-E group was significantly lower than that of the HFD-C group?P<0.05?.The shortened fraction?P<0.05?in the 4-2-A drosophila was significantly higher,and the shrink diameter?P<0.05?was significantly lower than that of the NF-C group in W-A.In 4-2-A drosophila,HFD-C group fibrous fibrillation?P<0.05?was significantly higher,and the shortened fraction?P<0.05?and diastolic diameter?P<0.05?were significantly lower than those of NF-C.The professed fibrillation?P<0.05?in HFD-E group was less significant,and the shortened fraction?P<0.05?and diastolic diameter?P<0.05?were significantly higher than the HFD-C group.3.3 PCR indicator test resultsIn W-A drosophila,the expression levels of NF-E group dfoxo?P<0.01?,Sod2?P<0.05?,Bmm?P<0.01?mRNA were significantly higher than in the NF-C group,and the expression level of dfoxo ?P<0.01?,Sod2?P<0.05?,and Bmm?P<0.05?were significantly lower.The level of expression of HFD-E group dfoxo?P<0.01?,Sod2?P<0.01?,Bmm?P<0.01?mRNA was significantly higher than that of HFD-C group.In 4-drosophila,the expression levels of HFD-C group Sod2 ?P<0.01?and Bmm?P<0.01?were significantly lower than in the NF-C group.The expression levels of HFD-E group Sod2?P<0.01?and Bmm?P<0.05?mRNA were significantly higher than in HFD-C groups.In 4-2-A drosophila,the expression levels of HFD-C group Sod2?P<0.01?and Bmm?P<0.05?mRNA were significantly lower than those of NF-C.The expression levels of HFD-E group Sod2?P<0.01?and Bmm?P<0.01?mRNA were significantly higher than in HFD-C group.4 Conclusion:1.A long-term high fat diet led to an increase in the body TAG level of drosophila,reduced body dfoxo,Sod2,Bmm mRNA expression,so that the drosophila diastoca diameter decreased and resulted in a reduction in the score of diastoic dysfunction,increased fibrous fibrillation.2.In high fat cases,regular exercise can improve body dfoxo,Sod2,Bmm mRNA expression level,reduce body TAG level and improve diastoic dysfunction and fibrillation caused by high fat poisoning.3.Regular exercise through the FOXO/SOD2 pathway to properly improve high fat diet caused by heart dysfunction,fibrous fibrillation.
