The aim of study:To observe the effect of high-intensity interval training on autophagy-related protein and mitochondrial complex protein expression in middle-aged mice,and to explore the effect of high-intensity interval training on hippocampal plasticity of middle-aged mice,and provide theoretical and experimental basis for scientific exercise of middle-aged people.Methods:Thirteen healthy male ICR mice,12 months old,were randomly divided into control group(CON,n=6)and high-intensity interval training group(HIIT,n=7).Among them,the CON group mice did not have any form of exercise intervention.The HIIT group mice were first subjected to 2 weeks of exercise preconditioning,and then the maximum endurance test was performed.According to the test results,a high-intensity interval training program was developed,and the exercise intensity at high intensity was 85%-90%of the maximum endurance,and the exercise intensity at the interval was 50%-60%of the maximum endurance,5 days a week(rest on Wednesdays and Sundays)for 7 weeks.The spontaneous activity and inquiry behavior of the mice were tested by open field experiments.Western blot analysis was used to determine the expression of cAMP response element binding protein(CREB),brain derived neurotrophic factor(BDNF),microtubule-associated protein 1 light chain 3(LC3/MAP1LC3),sequestosomel(p62/SQSTM1),BCL2/adenovirus E1B 19 kDa interacting protein 3-like(Bnip3L)and mitochondrial complex(OXPHOS)proteins in hippocampus of mice.Results:1.The results of the Open field test showed that:Compared with the CON group,the number of entries into the central area in the HIIT group was significantly reduced(P<0.05),and the latency to enter the central part was significantly shortened(P<0.01).2.Western blot results showed that:(1)The expression of CREB and p-CREB in hippocampus total lysate of two groups of miceCompared with CON group,there was no significant change in CREB expression in hippocampus of HIIT group(P>0.05),but p-CREB expression was significantly decreased(P<0.05).(2)The expression of BDNF in hippocampus total lysate of two groups of miceCompared with the CON group,the expression of BDNF in the hippocampus of HIIT group was significantly decreased(P<0.01).(3)The expression of OXPHOS in hippocampus total lysate of two groups of miceCompared with CON group,the expression of mitochondrial complex Complex? in hippocampus of HIIT group was significantly decreased(P<0.05),and there was no significant change in the expression of mitochondrial Complex ?.The expression of mitochondrial Complex ?/Complex ?/Complex ?(P<0.05,P<0.05,P<0.01)was significantly increased.(4)The expression of LC3 in hippocampus total lysate of two groups of miceCompared with the CON group,the ratio of LC3 ?/LC3 ? in the hippocampus of the HIIT group decreased after 7 weeks of HIIT training,but the difference did not reach significant(P=0.0831).The expression of LC3 ?/?-actin also showed a downward trend,but the difference was not significant(P=0.0943).(5)The expression of p62 in hippocampus total lysate of two groups of miceCompared with CON group,there was no significant difference in p62 protein expression in hippocampus of HIIT group(P>0.05).(6)The expression of LC3 in hippocampus mitochondrial lysate of two groups of miceCompared with the CON group,the ratio of LC3 ?/LC3 ? in the hippocampal mitochondrial lysate of the HIIT group was significantly decreased(P<0.01),and the expression of LC3 ?/VDAC was also significantly decreased(P<0.01).(7)The expression of p62 in hippocampus mitochondrial lysate of two groups of miceCompared with CON group,there was no significant change in p62 expression in hippocampal mitochondrial lysate of HIIT group(P>0.05).(8)The expression of Bnip3L in hippocampus mitochondrial lysate of two groups of miceCompared with the CON group,the expression of Bnip3L in the hippocampal mitochondrial lysate of HIIT group was significantly decreased(P<0.01),Conclusions:1.High-intensity interval training reduces the spontaneous activity and exploration behavior of middle-aged mice.2.High-intensity interval training inhibited CREB-BDNF signaling pathway in hippocampus of middle-aged mice.3.High-intensity interval training down-regulated the protein expression of mitophagy and Complex ? in hippocampus of middle-aged mice,and up-regulated the protein expression of Complex ?,? and ?.4.The behavior of mice in high-intensity interval training group may be related to the regulation of mitophagy and mitochondrial complex by high-intensity interval training.
