Objective: To research the effect of exercise and HIF-1? high-expression on the Nrf2 and its target genes in the skeletal muscle of mice.Methods: The C57BL/ 6J wide mice and HIF-1? high-expression mice were divided into 2 groups randomly.Control group and exercise for 3 hour.The exercised mice were subject to a moter-driven treadmill for 3 hours at a speed of 14m/min with a slope of 10%.The mice were sacrificed immediately after the exercise.The expression of 4-HNE?Keap1?Nrf2 and its target genes in skeletal muscle was analyzed using Western blot.Result:(1)After 3hours exercise,the expression of Nrf2 in nucleus of wild mice have significantly decreased,the expression of 4-HNE?Keap1 and HO-1 in cytoplasm have significantly decreased.(2)In rest state,compare to the wild mice,the expression of 4-HNE in cytoplasm of HIF-1? high-expression mice were significant decreased,the expression of SOD2 in cytoplasm of HIF-1? high-expression mice have significantly increased.(3)After 3 hours exercise,the expression of Nrf2 of HIF-1? high-expression mice was significantly increased,the expression of Keap1 in cytoplasm of HIF-1? high-expression mice was significantly increased,the expression of 4-HNE?NQO1 and SOD2 in cytoplasm of HIF-1? high-expression mice were significantly increased.Conclusion:(1)In rest state,the HIF-1? high expression can raise the antioxidant effective.(2)After 3hours exercise,the HIF-1? high expression have positive affections to the expression of Nrf2.
