Background:Peanut stems and leaves have been used,as medicinal materials to treat insomnia since ancient times,but the active ingredients in them are complex.No detailed research has been done,Some studies have found that w ater extracts of peanut stems and leaves contain four main chemical sub stances Alcohols,5-hydroxy-4',7-dimethoxyflavone,isoliquiritigenin,and ferulic acid):which may be active substances that prodnce sleep-promoting effects.Thi s article further studied the effect of isoliquiritigeninon sleep promotion,sleep mechanism and changes of sleep phase.OBJECTIVE:To investigate the effects of peanut stalk extract isoliquiritigenin(ILTG)on the sleep of C57 mice and the possible brain regions of its drug effects.Methods:Two-month-old C57 mice were tested.The mice were first surgically placed with a sleep recording device.The control group mice were perfused with physiological saline,and the administration group mice were perfused with isoliquiritigenin(32.4 mg/kg).Then,the mice were subjected to EEG,EMG recording and software to analyze the sleep and wake processes of mice;a water maze experiment was used to investigate whether isoliquiritigenin damaged the memory ability of mice;to observe the whole brain of mice C-fos staining was performed to observe the area of the brain where the drug was acting.Results:(1)On the 7th day of miragastric administration,the total amount of sleep of the administration group was significantly increased compared to the control group;among them,the administiation group and non-rapid eye movement(NREM,non-rapid eye movement))There was a significant increase and no significant difference in rapid eye movement(REM,rapid eye movement);on the 7th day of administration,the sleep latency of the administration group was significantly shortened compared with the control group,and the brain electrical energy spectrum was also compared to the control group.Significantly increased;(2)In the water maze experim ent,there was no significant difference in the time to find the platfora area(fifth quadrant)between the control group and the adminstumtien group,and there was no sigrificant dfference in the residence time in the platform area;(3)About one how after administration,the brain areas activated by isoliquiritigenin were ventrolateral preoptic area(VIPO)Abdominal median anterior visual nucleus(VMPO)and inferior vault(SFO);and isoliquiritigenin also increased the sleep deration of Fmr1 KO mice.Conclusion:ILTG car shorten the latency of C57 mice to fall asleep and increas e the amount of NREM sleep,and at the same time increase the energy spectrum of brain waves during NREM.The brain regions activated by the sleep promoitng effect of ILTG include VLPO,VMPO and SFO brain regions,and Isoglycyrrhizin has no neurotoxicity and will not damage the cognition and memory ofmice,and it can also improve congenital sleep disorders.
