Objective: To study the effects of receptor for advanced glycation end products(RAGE)on lipopolysaccharide-mediated expression of matrix metalloproteinase-2(MMP-2)and matrix metalloproteinase-9(MMP-9)in A549 cells.Methods: Using optimized dose of LPS(10?g / ml)to stimulate alveolar epithelial A549 cells,quantitative real-time polymerase chain reaction(q RT-PCR)and Western blot were used to detect the expression of MMP-2 and MMP-9.Detection of MMP-2 and MMP-9 protein secretion levels in A549 cells by ELISA.Construction of exogenous si RNA interferes with the expression level of RAGE in A549 cells,combined with the use of signal transduction pathway inhibitors,the effects of RAGE knockdown on LPS-induced MMP-2,MMP-9 expression and activation of NF-?B signaling pathway were detected by Western blot.Results:(1)LPS mediates the expression of MMP-2 and MMP-9 in A549 cells(P <0.05).(2)LPS mediates the expression of MMP-2 and MMP-9 by activating the NF-?B signaling pathway(P <0.05).(3)si RNA knocked down RAGE A549 cells,inhibited the NF-?B signaling pathway,and reduced the expression of MMP-2 and MMP-9(P <0.05).Conclusion: LPS mediates the expression of MMP-2 and MMP-9 in A549 cells through RAGE and NF-?B signal transduction.
