Objective1.Using the endogenous differential mi RNA(micro RNA-26 b,micro RNA-138,micro RNA-206 and micro RNA-let-7e)found in Prolactin-type pituitary adenomas(PRL-type pituitary adenomas)and Growth hormone type pituitary adenomas(GH-type pituitary adenomas)as detection of gene(genes for detection)aims to compare the expression levels of micro RNAs in nonfunctional pituitary adenoma subjects and healthy controls,and to explore the similarities and differences of the expressions of mi RNA between nonfunctional pituitary adenoma and PRL-type pituitary adenomas and GH-type pituitary adenomas.2.Comparing the expression level of mi RNA in non-functional pituitary adenoma subjects before and after "Chuining Decoction" treatment,and combining with the evaluation index of clinical efficacy to explore the mechanism of "Chuining Decoction" in the treatment of non-functional pituitary adenoma.Method Ten untreated non-functioning pituitary adenomas volunteers intended to take "Chuining Decoction" for one year were selected as subjects in this study,and 10 normal subjects were selected as controls.Peripheral blood samples were collected and four differentially expressed mi RNAs(micro RNA-26 b,micro RNA-138,micro RNA-206 and micro RNA-let-7e)obtained in the earlier stage of the test group were detected,recorded and archived by qrt PCR.By comparing the expressions of mi RNA and target gene mi RNA before and after the treatment,as well as the expression level of mi RNA related to normal people before and after the treatment,the difference and significance of endogenous mi RNA expressions were explored through statistical analysis.Result1.Before the treatment,the expressions of micro RNA-26 b,micro RNA-138,micro RNA-206 and micro RNA-let-7e in peripheral serum of NFPAs subjects were significantly different from those of normal subjects(P < 0.05).2.One year after the treatment of Pituitary Adenoma with "Chuining Decoction",the scores of TCM syndromes were evaluated.6 cases were effective,4 cases were markedly effective.Sellar region of 3 subjects showed that the tumors shrank.Seven patients showed no significant changes in the tumors.The expressions of micro RNA-138 and micro RNA-let-7e were significantly up-regulated,and there was no significant difference between micro RNA-138 and healthy subjects(P>0.05).Conclusion1.mi R-26 b,mi R-138,mi R-206 and mi R-let-7e participated in the occurrence and development of nonfunctional pituitary adenomas,and the expression of mi RNA was consistent with those of PRL-type pituitary adenomas and GH-type pituitary adenomas,which provided a basis for further exploration of the tumor markers of pituitary adenomas.2."Chuining Decoction" has a remarkable effect in the treatment of nonfunctional pituitary adenomas,and has certain regulating effect on the differential expression of mi RNA.mi R-138 and mi R-let-7e are obviously up-regulated.Among them,after treatment,the level of mi R-138 in subjects tends to apparel to the level of normal people.In this way,it provides some ideas for further exploration on the mechanism of Pitulining Prescription regulating endogenous differential mi RNA in various types of pituitary adenomas.The effective treatment of non-functional pituitary adenoma with traditional Chinese medicine provides laboratory evidence.
