| 【Object】The mouse model of asthma was induced by BALB/c mouse OVA,and the intervention of Sanzi Yangqin Decoction and dexamethasone was used to observe the airway resistance of Sanzi Yangqin Decoction in asthma model,pathology,total white blood cell count and lung tissue in BLAF.In the impact of inflammatory factors and other aspects,to explore its effectiveness and effective concentration,and to find possible targets of Sanzi Yangqin Decoction,to provide a basis for later research.【Method】First,21 BALB/c female mice were divided into three groups(blank group Con,nebulized group AI,and nasal group ND)for modeling experiments.The pathological changes of left lung tissue(HE staining)were observed in each group.Changes in total leukocyte counts in BALF and changes in TNF-α and INF-γ factors in the right lung tissue(ELISA)were used to determine animal modeling methods.In the later stage,78 BALB/c female mice of 8 weeks old were used as subjects,and were randomly divided into 6 groups.They were blank control group(Con),asthma model(A),Sanzi Yangqin Decoction high-dose treatment group(SH),medium-dose treatment group(SM),low-dose treatment group(SL)and dexamethasone treatment.Group(DXMS).Twelve groups in each group were adapted for 10 days in a clean animal room.Mice(Group A,SH,SM,SL,and DXMS groups)were sensitized by intraperitoneal injection(ip)A in the morning of 1 and 14 days,and in the afternoon after the second sensitization,nose(nd)stimulation..After ip anesthesia on the 25 th to 27 th day,continuous B droplets were nasal for 3 days.The sensitization and challenge of the blank control group(Con)were replaced by0.9% Na Cl solution.The time and dose were the same as those of the other groups.On the 28 th day,mice in each group began a one-week intervention treatment with three concentrations of Sanzi Yangqin Decoction(24 mg/ml),medium(12 mg/ml),and low(6 mg/ml).The Chinese medicine group(SH,SM,SL)mice were intragastrically administered daily,and DXMS group mice were administered ip at a dose of 1 mg/kg/d.On the 35 th day,the airway resistance value was measured and themice were sacrificed.The blood was used to detect the serum Ig E concentration,the alveolar lavage fluid was taken to determine the total number of white blood cells and the eosinophil ratio,and the left lung tissue pathological section was observed.The content of TNF-α,IL-2,IL-6,IFN-γ,IL-12p70,IL-4,IL-5 and IL-13 in the homogenate was measured by ELISA after the automatic sample grinder was ground.Therapeutic effect of Sanzi yangqin Decoction on bronchial asthma.Human airway epithelial NCI-H292 cells were used in vitro study,with control group(Con),EGF group(EGF,50 ng/ml),Sanzi Yangqin decoction group(SZYQT,1mg/ml),and EGF plus Sanzi Yangqin decoction group(EGF+SZYQT,the same concentration as the previous two groups),the expression level of Muc5 ac was detected by fluorescent quantitative PCR after 24 hours of drug administration.【Result】In this experiment,the appearance and behavior of the mice in each group were observed.The asthma model group(A)had obvious symptoms of scratching the nose and breathing deeply,and the other OVA sensitized groups were all relieved to varying degrees.Among them,Sanzi Yangqing Decoction high dose group(SH)After 7 days of treatment,the appearance was not significantly different from the control group,and the control group(Con)mice had no obvious symptoms.Experimental studies on the total cell count of alveolar lavage fluid and the percentage of total cells in EOS have shown that the application of high and medium doses of Sanzi Yangqin Decoction can significantly reduce the number of inflammatory cells in the airway and alveoli of asthmatic mouse models(P <0.05).The results of serum Ig E test showed that the application of high dose of Sanzi Yangqin Decoction can reduce the serum Ig E content in asthmatic mice model,and there is no statistical significance between the two groups(P>0.05).),demonstrating that there is no difference in the effect of the two doses on serum Ig E levels.And the experimental results show that the high dose of Sanzi Yangqin Decoction can reduce the degree of airway hyperplasia and improve the airway and alveolar disorders.Airway resistance test results showed that high doses in decoction can effectively reduce the airway resistance of asthmatic mice(P<0.05).In addition to IL-2(P>0.05),the inflammatory factors of lung tissue in the middle and high dose groups of Chinese medicine were decreased to different degrees.【Conclusion】1.Sanzi Yangqin Decoction can effectively reduce the serum Ig E content of asthmatic mice and the number of inflammatory cells in mouse BALF,and reduce the content of some inflammatory factors in lung tissue.2.Sanzi Yangqin Decoction can significantly reduce airway resistance in asthmatic mouse models.3.The high and dose application of Sanzi Yangqin Decoction has a certain therapeutic effect on bronchial asthma.4.Sanzi Yangqin Decoction can reduce the high expression of Muc5 ac of EGF-induced NCI H292 human airway epithelial cells. |
