Study On The Intervention Effect Of Shenge Powder On Doxorubicin-induced Heart Failure In Rats With Dilated Cardiomyopathy

Objective: To investigate whether Shenge Powder can improve the cardiac function of rats with dilated cardiomyopathy and heart failure;to explore the mechanism of Shenge Powder on the intervention of doxorubicin-induced heart failure in rats with dilated cardiomyopathy.Methods: 40 rats were randomly divided into normal group(n=10),model group(n=30).A rat model of dilated cardiomyopathy heart failure was established by intraperitoneal injection of doxorubicin 2.5 mg/kg once a week for 4 weeks.After successful modeling,model rats were randomly divided into control group,captopril group and Shenge powder group(n=10).The captopril group was given captopril at 5 mg/kg/day,and the Shenge powder group was intragastrically administered at 1.89 g/kg/day.The normal group and the control group were given distilled water at 10 ml/kg/day for 4 weeks.After 4 weeks,the rats underwent echocardiography,and the left ventricular end-diastolic diameter(LVIDd),left ventricular end-systolic diameter(LVIDs),left ventricular ejection fraction(LVEF),and left ventricular short-axis shortening rate were recorded.(FS),detection of serum BNP,cTnI indicators.Myocardial histopathological sections were observed.The m RNA and protein expression levels of SERCA2 a,PLB,cTnT and Cx43 in rat myocardial tissue were determined by RT-PCR and Western-blot.Results:1.Cardiac ultrasound results: Compared with the normal group,the LVIDd and LVIDs in the control group increased,LVEF and FS decreased,P<0.05;the LVIDd and LVIDs decreased in the captopril group and the Shenge group compared with the control group.LVEF,FS increased,P<0.05.2.Serum BNP: The control group was significantly higher than the normal group;the BNP was lower in the captopril group and the Shenge group than in the control group,P<0.05.3.Serum cTnI: The control group was significantly higher than the normal group;the captopril group and the Shenge powder group decreased compared with the control group,P<0.05.The captopril group was compared with the Shenge group,P>0.05.4.Myocardial histopathological changes:4.1 HE staining of myocardial tissue: the morphology and structure of normal myocardial cells were normal;intact nuclei could be seen,and there was no edema in the intercellular space.The myocardial cells in the control group were abnormal in morphology and myocardial fibrosis was obvious.The nucleus shrinks or deforms or even disappears.There is edema and widening in the intercellular space.The captopril group and the Shenge powder group showed slight improvement compared with the control group.4.2 Masson staining of cardiomyocytes: There were some collagen tissues around the normal group of cardiomyocytes.Some of the myocardium in the control group,Shenge powder group and captopril group had been replaced by collagen tissue.The captopril group and the Shenge powder group were between Between the normal group and the control group.5.SERCA2 a,PLB,cTnT,and Cx43 m RNA expression levels in rat myocardium: compared with the control group,SERCA2 a,Cx43 m RNA was increased in normal group,captopril group and Shenge group,P<0.05;PLB,cTnTm RNA were all Reduced,p < 0.05.6.SERCA2 a,PLB,cTnT,Cx43 protein expression levels in rat myocardium: SERCA2 a,Cx43 protein levels were increased in normal group,captopril group and Shenge group compared with the control group,P<0.05;PLB,cTnT protein Both decreased,p < 0.05.Conclusion:1.Shenge Powder has a certain effect on the reduction of the inner diameter of the heart cavity in rats with dilated cardiomyopathy and heart failure.It also has the function of increasing ejection fraction and lowering serum BNP and cTnI.2.Shenge Powder can also regulate the m RNA and protein expression of myocardial tissue SERCA2 a,PLB,cTnT and Cx43.
