Objective: According to the relevant records of yam,cortex phellodendron,and ginkgo in the literature of the past dynasties,the yam,cortex phellodendron,and ginkgo of the famous classical prescription Yihuang decoction group were tested;The quality evaluation of cortex phellodendron and ginkgo was studied by HPLC.It provides some reference for the research and application of a classical prescription Yihuang Decoction.Methods:1.A literature study on the origin of Chinese herbal medicines in the classic prescription Yi Huang DecoctionThrough consulting the herbal literature of ancient dynasties of relevant medicinal materials,sorting and screening the herbal literature of ancient dynasties,analyzing the proper names of the medicinal materials,the morphology of the original plants,the distribution of the origin of the medicinal materials recorded in the literature of past dynasties,and synthetically verifying the origins of,yam,cortex phellodendron and ginkgo,which are the classical prescriptions of Yihuang decoction.2.Study on quality inspection of traditional Chinese medicines of a classical prescription Yihuang decoction granuleAccording to the requirements of the General Principles of the First and Fourth Editions of the Chinese Pharmacopoeia(2015 Edition),the five herbal medicines Yam,Radix Scutellariae,Cortex Phellodendri,Fructus Plantagonis and Fructus Alba in the prescriptions of Yihuangtang Granule were examined.The items included character,TLC identification,content determination,moisture determination,extract determination,ash content and acid-insoluble ash content determination.3.Study on fingerprint of cortex phellodendron,cluster analysis and principle component analysis of Cortex PhellodendriThe fingerprints of Cortex Phellodendri were established by HPLC and the relative retention time and peak area were used as the index for methodological investigation.The fingerprints of 19 samples of Cortex Phellodendri were examined and analyzed by HPLC and the common model was established with Phellodendron hydrochloride and berberine hydrochloride as reference.The similarity of fingerprints of 19 batches ofCortex Phellodendri was calculated by using the software of "Similarity Evaluation System of Chromatographic Fingerprints of Traditional Chinese Medicine" in 2012 edition,and the similarity range of Cortex Phellodendri fingerprints was determined.The origin of Phellodendron amurense was determined by cluster analysis and principal component analysis of different content of Phellodendron amurense by SPSS21.0software..4.Study on the toxic constituents of ginkgolic acid in ginkgo(limit examination)The limit of total ginkgolic acid,a toxic component in ginkgo,was determined by HPLC.The limit of detection and quantitative limit of total ginkgolic acid in ginkgo were determined,and the method of limit detection was validated.According to the determination results of total ginkgolic acid in 15 batches of white fruits from 6producing areas,the toxic components of ginkgolic acid were evaluated comprehensively.5.Study on establishment of HPLC fingerprint,cluster analysis and principle component analysis of ginkgo.The fingerprint of ginkgo was established by HPLC.The relative retention time and relative peak area were used as indicators for methodological investigation.The fingerprints of 15 batches ginkgo medicinal materials were examined and analyzed by HPLC,and the common model was established with the reference of total ginkgolic acid A and total ginkgolic acid B.The similarity of fingerprints of 15 batches of ginkgos was calculated by using the software of "Similarity Evaluation System of Chromatographic Fingerprints of Traditional Chinese Medicine" in 2012 edition,and the similarity range of fingerprints of Cortex Phellodendri was determined.The origin of Cortex Phellodendri was determined by cluster analysis and principal component analysis.Result:1.A literature study on the origin of Chinese herbal medicines in the classic prescription Yi Huang DecoctionThe name,origin,origin,harvesting time and the historic changes of medicinal parts of Chinese yam,Cortex Phellodendri and the ginkgo medicinal material were determined to provide some reference for the follow-up study..2.Study on quality inspection of traditional Chinese medicines of Yihuang decoction GranuleAll original medicinal materials of the Five Herbs in the prescription conform to the inspection stipulations of the first and fourth parts of the Chinese Pharmacopoeia in the 2015 edition.The results of the inspection conform to the stipulations and can be used for follow-up research.3.Study on establishment of HPLC fingerprint,cluster analysis and principle component analysis of Cortex PhellodendriFingerprint determination method has good precision,repeatability and durability.The solution is stable within 24 hours.Six common peaks have been established,and the correlation coefficient is between 0.993 and 1.000.The origin of Cortex Phellodendri was determined to be Dayi County and Dujiangyan County,Sichuan Province.4.Study on the toxic constituents of ginkgolic acid in ginkgo(limit examination)The determination method of total ginkgolic acid in ginkgo was determined.The number of theoretical plates was more than 1000 and the separation of chromatographic peaks was good.After methodological investigation,the quantitative limit was 0.1046ug/m L,the detection limit was 0.02614 ug/m L,and the linear relationship was good in the range of 0.09970 ug/m L to 0.4985 ug/m L.The instrument had good precision,good repeatability,high accuracy and good durability.The solution was stable within 24 hours.The method was simple and feasible.According to the determination results of total ginkgolic acid in 19 batches of ginkgolic acid and comprehensive evaluation,it was found that there was a great difference in the toxic components of ginkgo from different habitats5.Study on establishment of HPLC fingerprint,cluster analysis and principle component analysis of ginkgo.Fingerprint determination method has good precision,repeatability and durability.The solution is stable within 24 hours.Six common peaks have been established with correlation coefficients ranging from 0.933 to 0.988.The origin of ginkgo medicinal material was determined to be Tancheng County and Yishui County in ShandongProvince.Conclusion:1.Based on the textual research of the original medicinal materials,it is determined that yam is the dried rhizome of Dioscorea opposita Thunb,a dioscoreaceous plant.The genuine producing areas are Wenxian County,Henan Province,Wuzhi County and other place;Cortex Phellodendri is the dry bark of Rutaceae yellow bark tree,and its genuine producing area is Sichuan;Gingko is the dried mature seed of Ginkgo biloba L.and its main producing areas are Shandong,Anhui and Jiangsu.2.The fingerprint of ginkgo and Cortex Phellodendri showed that there were differences in the components of medicinal materials from different origins.The fingerprint had obvious identification characteristics and could be used to identify the authenticity of medicinal materials.The test method is simple in operation,with little interference and high accuracy in judgment.It is feasible to control the quality of medicinal materials.It embodies the uniqueness of species chromatographic characteristics emphasized by the fingerprint of HPLC and the similarity between the same species.3.Through the study of total ginkgolic acid,the toxic component of white fruit was found to exceed the requirement of Chinese Pharmacopoeia 2015 edition(total ginkgolic acid is less than 10 ppm).Therefore,it is necessary to establish a method for the analysis of ginkgolic acid in white fruit to provide a basis for quality control and safety evaluation of white fruit products.
