Studies On Chemical Constituents Of Urceola Rosea,an Ethnic Medicine

In this paper,the chemical constituents were studied in the folk medicinal plantUrceola rosea?Hook.et Arn.?D.J.Middleton=Ecdysanthera rosea Hook.et Arn.Urceola rosea is a genus of the Apocynaceae family,which is used to treat rheumatism,bone pain,chronic nephritis and bruises and other symptoms.The Vietnamese Yao people use it to resist inflammation and infection,while the Jing people use it to treat chronic nephritis caused by kidney stones.Modern research shows that Urceola rosea has anti-fungal effect.In addition,in Yunnan,the plant is also a sour wild vegetable,which can be fried,and is very popular with local people.In this study,95%ethanol extract of Urceola rosea was extracted with petroleumether and ethyl acetate respectively.Sephadex LH-20,MCI,ODS,silica gel,HW-40F,and HPLC were used to separate the ethyl acetate fraction.The isolated compounds were identified by spectral analysis(ESI-MS,¹H NMR,¹³C NMR,HSQC,HMBC,NOESY,¹H-¹H COSY,IR,UV,CD/ORD,etc.)and combined with literature comparison.A total of 54 compounds were identified,including 34 steroids?glycosides?,18 lignans and two triterpenoids.Among them,there were 28 new compounds,including 22 C₂₁ steroids?glycosides?and six lignans.The structure of C₂₁ steroids?glycosides?on the side chain of D ring is significantly different from that of known steroidal glycosides,that is,a lactone ring E is formed on the basis of D ring,which has a new structure type;Aglycones,by linking different types of sugar chains,increase the novelty of these compounds.The structures of C₂₁ steroids?glycosides?new compounds 1,21,22 are shown as follows:.We used CCK-8 assay to test the effects of the isolated compounds on the inflammatory cell model of RAW 264.7 induced by LPS,and measure the inhibitory effects of them on TNF-?,IL-6 and the release of NO.Several compounds had anti-inflammatory activities,among them,compound 39 showed better inhibitory effects on IL-6 and NO release,and its IC₅₀ value was below 5?M,respectively.These studies not only enable us to understand the general situation of chemicalconstituents of Urceola rosea,but also have important significance for improving the chemical constituent types of Urceola rose,and lay a foundation for the study of its pharmacodynamic substances,which is conducive to exploring the mechanism of its pharmacodynamic effect.
