Object To investigate the expression of long noncoding RNA(Lnc RNAs)HOTAIR in acute myeloid leukemia(AML)and molecular mechanism of resistance to daunorubicin(DNR)in AML cells Method Collection 50 bone marrow mononuclear cells(AML primary cells)from AML patients in the Department of Hematology of the First Affiliated Hospital of Wenzhou Medical University,10 cases of bone marrow mononuclear cells from normal donors,and preserve AML in the internal medicine laboratory of the First Affiliated Hospital of Wenzhou Medical University Cell lines(HL60,THP1,U937,KG1,Kasumi-1,NB4,HEL,Kasumi-3,U937,THP1)and 293 T cell lines.With normal donor bone marrow mononuclear cells as the control group,q RT-PCR was used to detect the expression of HOTAIR in AML primary cells,AML cell lines,and normal control groups.Select THP1 cells and U937 cells for further study,construct sh-HOTAIR and sh-NC,and transfect the above two cells respectively.The transfected cells were treated with a final concentration of 0.1?mol/L daunorubicin(DNR),and the cell proliferation was detected by CCK8 experiment.Flow cytometry was used to detect the apoptosis of transfected cells.q RT-PCR was used to detect the expression of HOTAIR in the transfected cells,and western blot was used to detect the P-glycoprotein of the transfected cells.Result This study found that the q RT-PCR test results showed that the expression of HOTAIR in AML primary cells and AML cell lines was significantly higher than that of the normal control group;the results of CCK8 showed that the expression of HOTAIRwas down-regulated and cell proliferation was slowed down.The flow cytometry results showed that the apoptosis rate of down-regulated HOTAIR expression cells increased,and the down-regulated HOTAIR expression combined with DNR effect had a higher apoptosis rate than the cells that only down-regulated HOTAIR or only used drugs.Western blot results showed that the expression of HOTAIR was down-regulated,and the expression of P-glycoprotein in the cell was also down-regulated.Conclusion 1.HOTAIR is highly expressed in patients with acute myeloid leukemia and AML cell lines;2.Abnormally expressed HOTAIR may be closely related to the proliferation and apoptosis of acute myeloid leukemia cells;3.Abnormally expressed HOTAIR may regulate P-gp expression affects the resistance of AML cells to daunorubicin;4.The results of this study may provide new ideas and clues for the resistance of acute myeloid leukemia to daunorubicin.
