Objective: To detect the expression level of Dickkopf-1(DKK-1)in serum of patients with alcoholic osteonecrosis of the femoral head(Alcoholic osteonecrosis of the femoral head,AONFH)and the expression levels of DKK-1 and the downstream factors of Wnt signal pathway ?-catenin and phosphorylated ? / catenin(P-? / catenin)in bone tissue,to explore the relationship between them and AONFH,so as to provide a new idea for studying the pathogenesis of AONFH.Methods: In experiment 1,the sera of 43 patients with clinically diagnosed AONFH and 43 healthy subjects matched with sex and age were collected,and the relevant clinical data were recorded,and the expression level of Dkk-1 was detected by enzyme-linked immunosorbent assay(Enzyme-linked immunosorbent assay,ELISA).(2)according to the(Association Research circulation osseous,ARCO)staging of the International Society of Bone Circulation,the AONFH group was divided into two groups: group A(n = 21)in stage ??? and group B(n = 22)in stage ?? IV,and the results were compared between groups.(3)SPSS20.0 statistical software for statistical analysis,the measurement data conformed to the normal distribution and used independent sample t-test after the test variance was uniform,and the variance was uneven using approximate t-test,expressed by mean(x)±standard deviation(s);any group of data showed skewness distribution,using u-test method,P< 0.05.There was statistical significance.In experiment 2,27 femoral head specimens of AONFH were collected as the experimental group,and the femoral head tissues of 27 patients with femoral neck fracture after hip arthroplasty were taken as the control group.The expressions of Dkk-1,? / catenin and P-? / catenin in the two groups were detected by immunohistochemistry.(3)SPSS20.0 statistical software for statistical analysis,the measurement data conformed to the normal distribution and used independent sample t-test after the test variance was uniform,the variance was uneven using the approximate t-test,expressed by the mean(x)±standard deviation(s);any group of data showed skewness distribution,using the u-test method.P <0.05 indicates that it is statistically significant.Results: In experiment 1,(1)the level of serum Dkk-1 expression in AONFH group(4250.56 ±2250.48 pg / ml)was significantly higher than that in normal control group(2645.37 ±1926.79 pg/ml)(P<0.05).(2)the level of Dkk-1 in stage ??? group(6045.52 ±1322.38 pg/ml)was significantly higher than that in stage ??? group(2370.12 ±1242.06 pg/ml).In experiment 2,The level of Dkk-1 in bone tissue in AONFH group(2.57±2.04)was significantly higher than that in femoral neck fracture control group(0.85±0.21),P<0.05.The levels of ? / catenin and P-? / catenin in bone tissue in AONFH group were significantly lower than those in femoral neck fracture control group(P<0.05).Conclusion: 1.This study shows that the serotonin levels of Dkk-1 in AONFH patients increase,the severity of AONFH may be associated with Dkk-1 concentrations,and Dkk-1 may become one of the potential reference indicators for early diagnosis and treatment of AONFH in the future.2.Abnormal expression of Dkk-1 in peripheral serum and bone tissue in Patients of AONFH may be related to the pathogenesis of AONFH.
