Objective:To investigate peripheral blood T lymphocyte subsets,B lymphocyte,NK cells and immune globulin expression levels change in before and after chemother apy of diffuse large B-cell lymphoma patients,and analyse the relationship between di-sease occurrence,development and treatment.Methods:Sixty patients who were diagnosed with DLBCL by pathological in The Affiliated Hospital of Youjiang Medical University for Nationalities between January2016 and October 2019 as the study group,and 60 healthy subjects as control group in the same period.The levels of T lymphocyte subsets,B lymphocytes and NK cells were determined in flow cytometric,The levels of immune globulin were determined in automatic biochemical analyzer.To analyze the clinical significance of T lymphoc-yte subsets,NK cells,B-lymphocytes and immune globulin levels change in the periph-eral blood of the control group and the study group before and after chemotherapy.Results:(1)The levels of T lymphocyte subsets(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cell with DLBCL patients before chemotherapy were significantly lower than the control group(P<0.05),and CD8~+levels were significantly higher than the control group(P<0.05).CD19~+B lymphocytes and Ig A,Ig G expression levels were slightly lower than the control group,and the expression level of Ig M was slightly higher than the control group(P>0.05).(2)The levels of T lymphocytes(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with DLBCL patients after chemotherapy increased compared with before chemotherapy(P<0.05),CD8~+levels decreased compared with before chemotherapy(P<0.05).CD19~+B lymphocytes and Ig A,Ig G,Ig M expression levels were slightly higher than before chemotherapy(P>0.05).(3)DLBCL patients with effective chemotherapy,the levels of T lymphocytes(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with after chemotherapy were significantly increased than before chemotherapy(P<0.05),and CD8~+levels significantly decreased compared with before chemotherapy(P<0.05).The levels of CD19~+B lymphocytes and Ig A,Ig G,Ig M were slightly higher than before chemotherapy(P>0.05).(4)DLBCL patients with ineffective chemotherapy,the levels of T lymphocytes(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with after chemotherapy were slightly higher than before chemotherapy(P>0.05),and CD8~+levels were slightly lower than before chemoth-erapy(P>0.05).CD19~+B lymphocytes and Ig A,Ig G,Ig M levels increased slightly compared with the before chemotherapy(P>0.05).(5)DLBCL patients with before chemotherapy,the levels of T lymphocytes(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with effective chemotherapy were slightly higher than ineffective chemotherapy(P<0.05),and CD8~+levels were slightly lower than ineffective chemotherapy(P<0.05).CD19~+B lymphocytes and Ig A,Ig G,Ig M levels increased slightly compared with ineffective group(P>0.05).(6)DLBCL patients with after chemotherapy,the levels of T lymphocytes(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with effective chemotherapy were significantly increased than ineffective chemotherapy(P<0.05),and CD8~+levels was significantly lower than ineffective group(P<0.05).CD19~+B lymphocytes and Ig A,Ig G,Ig M levels slightly increased compared with ineffective group(P>0.05).(7)Compared with the control group,the levels of T lymphocyte subsets(CD3~+,CD4~+,CD4~+/CD8~+ratio)and NK cells with DLBCL patients of effective after chemotherapy,although they were higher than before chemotherapy,but still lower than the control group(P<0.05),CD8~+levels was lower than before chemotherapy,and still higher than the control group(P<0.05),the B lymphocyte CD19~+and Ig A,Ig G,Ig M levels were slightly higher than the control group(P>0.05).(8)Compared with the control group,the levels of T lymphocyte subsets(CD3+,CD4+,CD4+/CD8+ratio)and NK cells with DLBCL patients of ineffective after chemo therapy were significantly lower than the control group(P<0.05),CD8+levels was sig nificantly higher than the control group(P<0.05),the B lymphocyte CD19~+and Ig A,Ig G,Ig M levels were slightly lower than the control group(P>0.05).Conclusion:(1)DLBCL patients exist immunosuppression in the body with mainly cellular immunosuppression,and inexact relationship with humoral immunity.(2)The expression levels of T lymphocyte subsets and NK cells in peripheral blood are closely related to the occurrence,development and clinical efficacy of DLBCL.Dynamic monitoring of the expression levels of peripheral blood T lymphocyte subsets(CD3~+,CD4~+,CD8~+,CD4~+/CD8~+ratio)and NK cells in DLBCL patients can be used as one of the indicators for monitoring immune function,disease status and prognosis.
