Objective:To detect the expression of Aurora Kinase B and Survivin protein of chromosome passenger complex(Chromosomal Passenger Complex,CPC)in colorectal cancer(Colorectal Cancer,CRC)and its correlation with colorectal cancer;To explore the effect of reducing Aurora-B on proliferation,invasion and migration of colorectal cancer cells and its application in colorectal cancer diagnosis and treatment.It plays a positive role in the diagnosis,development and prognosis of colorectal cancer.Methods:The subjects were 75 cases of colorectal cancer diagnosed by the pathology department of Affiliated Hospital of Guilin Medical University from August 2015 to October 2018.The expression of Aurora-B and Survivin in colorectal cancerwas detected by immunohistochemistry.By taking the normal colorectal tissue group as the control group,the relationship between the expression of proteins and the clinicopathological features of colorectal cancer was analyzed.Atotal of 95 samples were selected,including 75 colorectal cancerpatients and 20 normal tissue samples as the control group.All the specimens were fixed with neutral formalin,dehydrated and embedded in paraffin.The sections of specimens were4 um thick and were stained by immunohistochemistry.Based on the preliminaryexperimental results,combined with cell culture techniques and methods,cell proliferation experiment and cell scratch experiment were used to detect the effect of Aurora-B reducing on the proliferation and migrationof colorectal cancer cells to further clarify the role of Aurora-B and Survivin in the development of colorectal cancer.Results:1.The positive expression rates of Aurora-B and Survivin in colorectal cancer were significantly higher than that in normal colorectal mucosa.The difference was statistically significant(P<0.05).2.The expression of Aurora-B and Survivin in colorectal cancer was correlated with the depth of tumor invasion,tumor differentiation and lymph node metastasis(P< 0.05).3.Reduce the expression of Aurora-B can significantly inhibit the proliferation of colorectal cancer cells.4.After reducing the expression of Aurora-B,the migration ability of colorectal cancer cells has decreased significantly,and the scratch experiment showed that the healing rate of the tumor cell transfection group was significantly lower than that of the control group.Conclusions:1.Aurora-B and Survivin are closely related to the clinicopathological features of colorectal cancer and may be used as a reference index to evaluate the prognosis of patients with tumor.2.Reduce Aurora-B in colorectal cancer is closely related to the occurrence and development of colorectal cancer cells,which can be regarded as diagnostic markers and therapeutic targets in clinical studies of colorectal cancer.
