Aims: The diagnosis of SLE is not an accurate indicator,but a series of scores and the mechanisms underlying it remains only partially understood.The new perspective is needed to elucidate the pathophysiology of systemic lupus erythematosus development.So we explore whether transfer RNAs(tRNAs)and small RNAs derived from transfer RNA(tsRNA)contribute to SLE physiopathologic mechanism.Methods: Total RNA from peripheral blood mononuclear cells(PBMC)of 20 SLE patients and 20 normal controls(NCs)were extracted tsRNA and tRNA expression profiling was follow by Next Seq system.Target gene were predicted by informatics analysis.Subsequently,to explore the function of message RNA(m RNA)in this target gene,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed using Cytoscape plug-in Bin Go,DAVID database,and Cytoscape software.Results: 101 tRNAs were differentially expressed in SLE patients versus HCs by RNA microarray,including 50 tRNAs were significantly up-regulated and 51 were downregulated among the SLE samples.We also identified a total of 355 tsRNA as differentially expressed profiles.GO analysis revealed that the altered target genes of the selected tRNAs and tsRNAs most enriched similarly in immune response,immune system process.Moreover,KEGG pathway analysis demonstrated that altered target genes of tRNAs most enriched in Systemic lupus erythematosus.While the altered target genes of tsRNAs most enriched in T cell receptor signaling pathway,Th1 and Th2 cell differentiation and primary immunodeficiency.Which may have related with the initiation of SLE.Conclusion: Our results provide a novel perspective for studying tRNA-related and tsRNA-related pathogenesis in SLE.
