Objective: To overcome the problems of a single mechanism that the research on scarless wound healing,which was unable to meet the four complex biological processes in the healing of diabetic ulcers.This study proposed a concept of ordered diabetic ulcer recovery and designed 20(s)-protopanaxadiol nanostructured lipid carrier gel,due to the synergistic effect of the carrier in the inflammatory phase,the proliferative phase and the remodeling phase,achieving orderly recovery of the wound,and finally achieving scar-free healing.Methods: The particle sizes,drug-loading capacity and distribution in the silicone elastomer were investigated by particle size analyzer,dialysis bag,scanning electron microscope and freeze-scanning electron microscopy.The drug release behavior of the simulated carrier in the wound site was obtained by in vitro release.In vitro cell assay was used to detect the biological function of inflammatory cells and vascular endothelial cells,and a full-thickness wound model of diabetic mice(db/db)was established.The histopathological method was used to evaluate the inflammatory cell infiltration and proliferative phase of the wound during the inflammatory phase of the wound.Effects of collagen deposition during angiogenesis and remodeling.Results: Compared with the PPD solution group,the NO content in PPD-N and PPD-NS decreased significantly.In the cell migration experiment,the area of PPD-N and PPD-NS treatment group The extent of the reduction was even more pronounced.At 48 h,the scratch area of the PPD-NS group had basically healed,while the closure rate of the PPD solution group only reached 56.89 ± 5.81%;the number of vascular nodes in the PPD-N group increased significantly,especially in the PPD-NS group.The number of vascular nodes was the highest;the results of in vivo tissue section showed that compared with the PPD solution group,the number of inflammatory cells in the inflammatory phase of the PPD-NS group was significantly decreased,and the angiogenesis density in the proliferative phase was significantly increased.The remodeling collagen was arranged around the skin appendage.Conclusion: PPD-NS has good anti-inflammatory and pro-angiogenic ability in vitro,and due to the synergistic effect of PPD nano-lipid carrier and silicone elastomer,it could reduce the infiltration density of inflammatory cells in the inflammatory phase in vivo and promote the regeneration of angiogenesis during proliferative phase.Regulating collagen deposition during remodeling,the inflammatory phase,proliferative phase and remodeling phase of the wound were smoothly carried out,and the orderly recovery of diabetic ulcer was achieved,and finally the scar-free healing effect was achieved,which providing new ideas for the treatment of clinical diabetic ulcer.
