Objective: To investigate the effect of acupuncture preconditioning on the signaling pathway of rapamycin target protein(m TOR)in skeletal muscle of rats with heavy load training.Methods: 96 male SD rats were randomly divided into 3 groups: control group(Normal,N=32),Model group(M=32),and Acupuncture Pretreatment group(AP=32).No training and no acupuncture in group N,and only the same grasping action as in group AP;In the M group,1 time/day,6 times/week,a total of 8 weeks of continuous increasing heavy load treadmill running training;The AP group received the same training program as the M group after acupuncture on " ST36 and CV4 acupoints ",with acupuncture once per day,20 min per time and 6 times per week for a total of 8 weeks.The expressions of Akt,m TOR,p70s6 k,4E-BP1 and AMPK m RNA were detected by RT-PCR in 8 rats randomly selected from each group after 24 h of exercise or adjusted rest at week 2,5,8 and 9,respectively.Western blotting method was used to measure the expression of m TOR,p70s6 k and their phosphorylation.Results: 1.The weight of the rats in each group showed an increasing trend,but the high-load training inhibited the weight gain of the rats.After 1 to 2 weeks,the weight of the M group and the AP group was lower than that of the N group(P < 0.01,P < 0.05).After 4 weeks,M group was lower than N group(P < 0.05),and AP group was higher than M group(P < 0.05).After 5 weeks,both M group and AP group were lower than N group(P < 0.01,P < 0.05).After 6 weeks,M group was lower than N group(P < 0.01),and AP group was higher than M group(P < 0.01).After 7 weeks,body weight of M group was lower than that of N group(P < 0.01),and that of AP group was higher than that of M group(P < 0.05).After 8 weeks,both M group and AP group were lower than N group(P < 0.01,P < 0.01)..2.Changes of gastrocnemius muscle mass index: after 2week,AP group was lower than N group(P < 0.01);After 8week,the AP group was lower than the N group(P < 0.01).AP group was lower than M group(P < 0.05).3.Akt m RNA content: after 5week,the AP group was higher than the M group and the N group(P < 0.05,P<0.05).After 8week,group M was higher than group N(P < 0.05),and group AP was higher than group M and N(P < 0.05,P < 0.01).4.m TOR m RNA content: after 2week,the content of M group was higher than that of N group(P < 0.01),while that of AP group was lower than that of M group(P < 0.01).After 5week,the content of AP group was higher than that of M group and N group(P < 0.01,P < 0.01).After 8week,the content of M group was higher than that of N group(P < 0.01),and that of AP group was higher than that of M group and N group(P < 0.01,P < 0.01).5.p70s6 k m RNA content: after 2week,group M increased compared with group N(P < 0.01),while group AP decreased compared with group M(P < 0.01).After 5week,the AP group was higher than the M and N groups(P < 0.01,P < 0.01).After 8week,M group increased compared with N group(P < 0.01),and AP group increased compared with M group and N group(P < 0.01,P < 0.01).6.4E-BP1 m RNA content: after 2week,the M group was lower than the N group(P < 0.05),and the AP group was lower than the N group(P < 0.01);7.AMPK m RNA content: after 5week,the AP group was lower than the M group(P < 0.05).8.m TOR protein content and its phosphorylation(m TOR-ser2488): after 2week,5week and 8week,AP group was higher than N group(P < 0.01,P < 0.01,P < 0.05).;p-m TOR increased in the AP group compared with the M group(P < 0.01,P < 0.01,P < 0.01).9.p70s6 k protein content and its phosphorylation(Thr389): after 8week,P-p70s6 k increased in the AP group compared to the M group(P < 0.01).Conclusion: 1.8 weeks of high-load training increased the expression of Akt,m TOR,p70s6 k m RNA in the skeletal muscle of rats,which may be one of the mechanisms that high-load training promotes the improvement of exercise capacity.2.Compared with high-load training,acupuncture pretreatment further increased the expression of Akt,m TOR,and p70s6 k m RNA in the skeletal muscle of rats under 8-week high-load training,and led to increased phosphorylation of m TOR and its downstream signaling protein p70s6 k,and increased protein synthesis,suggesting that acupuncture pretreatment can further improve athletic ability compared with high-load training.
