Chinese medicine is a natural medicine with a certain pharmacological effect,which is a great tradition of the Chinese nation.The chemical composition of Chinese medicinal materials is unique and complex,with numerous modification sites and abundant sources,and their physical and chemical properties are also different.Among them,the main components of traditional Chinese medicine mainly include alkaloids,flavonoids,anthraquinones,volatile oils,terpenes,organic acids and sugars.In recent years,the rapid development of supramolecular chemistry has become a important subject in the 21 st century,and through the host-guest combination,the main body of supramolecular can be combined with the relevant monomer components of traditional Chinese medicine to change the poor solubility and poor bioavailability of the monomer components Shortcomings.Calix [n] arene(Cn A)is formed from phenol units through methylene bridges and is similar in shape to a class of cyclic compounds in Greek wine glasses.Through recent studies,the chemical structure of calixarene was modified,and the upper edge was modified to obtain sulfonate,forming sulfonated calixarene(p-Sulfonated calix [n] arene).Its preparation is simple,the main inclusion driving force is stronger in the aqueous phase,and its upper edge sulfonate enhances the electronegativity of the calixarene cavity,and provides it with additional electrostatic binding sites.The aqueous phase exhibits extremely strong bonding ability to organic cations.In order to study the binding effect of s p-Sulfonated calix [6] arene on traditional Chinese medicine components,this topic selected 28 traditional Chinese medicine monomers,which are commonly used active ingredients in traditional Chinese medicine ingredients,such as flavonoids,anthraquinones,alkaloids,and organic acids.From the perspective of in vitro biopharmaceutics,the effect of p-Sulfonated calix [6] arene on the monomeric components of traditional Chinese medicine was studied on the basis of the effect of p-Sulfonated calix [6] arene on the solubility of the monomeric components of traditional Chinese medicine.The binding constant was determined by fluorescence competitive titration to explore the mechanism of action,and it was further applied to the extraction and separation of the alkaloids of Chinese medicinal components.Finally,the p-Sulfonated calix [6] arene hydrocarbon was used as a carrier to study the release of guest molecules after drug loading.1.In this experiment,high-performance liquid chromatography(HPLC)was used to analyze the solubilization effect of p-Sulfonated calix [6] arene on the 28 Chinese medicine monomer components.Using peak area as an indicator,various methods of analysis were used to analyze solubilizing effect of the p-Sulfonated calix [6] arene.Fluorescence competition titration was used to determine the bonding constants of p-Sulfonated calix [6] arene and alkaloid monomer components,and the relationship between them and solubility was analyzed.The research results show that the p-Sulfonated calix [6] arene shows obvious solubilization effect on alkaloid monomers,but it does not show obvious solubilization effect on flavonoids,anthraquinones and aromatic organic acids,but on rhein it exhibits a solubility-reducing effect.Through the measurement of the bonding constant,it is found that the solubilization effect of most alkaloid monomers has a positive correlation with the bonding constant and is related to its own solubility.If the guest component has good solubility,the solubilization effect will be reduced.2.In this experiment,p-Sulfonated calix[6]arene was used as a supramolecular material in the extraction and separation of traditional Chinese medicine components.The research of p-Sulfonated calix[6]arene's extracting ability was conducted by using nuciferine as a drug model.The optimal conditions for p-Sulfonated calix[6]arene to extract nuciferine were determined by single factor investigation.It was determined that p-Sulfonated calix[6]arene was separated from nuciferine by adjusting the p H value.Good dissociation conditions.The experimental results found that the effect of solubilization and extraction of p-Sulfonated calix[6]arene was far more than that of cyclodextrin,and after the traditional water extraction method p-Sulfonated calix[6]arene was extracted,both the extraction amount and the purity phase were greatly increased.The best extraction conditions are cold immersion extraction,the extraction time is 1 h,the added material-liquid ratio is 1: 20,the lotus leaf using a No.5 sieve,p-Sulfonated calix[6]arene : lotus leaf alkali is 0.4: 1.Finally,the drug effect model of nuciferine and quinine was used to study the separation effect of p-Sulfonated calix[6]arene from flavonoids and anthraquinones.After mixing alkaloids with flavonoids and anthraquinones,p-Sulfonated calix[6]arene can separate alkaloids well through selective binding,and obtain a pure product through alkalization.3.In this experiment,quinine,nuciferine,and matrine were used as model drugs to study the encapsulation of alkaloid guest molecules by p-Sulfonated calix[6]arene as a drug carrier,and then release in vitro.Experiments have found that the release conditions of alkaloid monomers in inclusion complexes are related to their water solubility.The alkaloids(matrine)with better water solubility have similar in vitro release behavior to their monomer components after inclusion.For scopolamine,which has poor water solubility,the release effect after complexing with p-Sulfonated calix[6]arene at different p H is different from that of monomer,of which p H 7.4 is the most obvious.Under the condition of p H 7.4,the release of nuciferine @ p-Sulfonated calix[6] arene complex was the slowest,followed by p H 4.5 and the fastest release was p H 1.2.Through the study of this subject,the solubilization effect of p-Sulfonated calix[6]arene on a variety of traditional Chinese medicine monomer components was determined,and the interaction between p-Sulfonated calix[6]arene and alkaloid components was initially studied.It may have a solubilizing effect on the monomer components of Chinese medicine of alkaloids,and then explore its application research.It is desirable to introduce sulfonated calixarene into important areas.
