Objective:Organic anions and cations have been studied for the characteristics of Penicillium purpurogenum Li-3 and its catalytic application.Methods:1.Four different organic anions and cations salts were added to the medium to investigate the effects of organic anions?PF6-,Tf2N-?and organic cations?BMIM+,MBPy+?on the growth and catalytic activity of Penicillium purpurogenum Li-3.2.The methylimidazole cation?MIM+?is covalently attached to the nanosilica particles to obtain the nanosilica grafted methylimidazole?Si O2-MIM+?cation through organic synthesis.This cation is combined with hexafluorophosphate anion?PF6-?and bistrifluoromethanesulfonyl?Tf2N-?,respectively Imine anion to obtain the product nano-silica grafted ionic liquid.3.Sodium alginate?SA?and polyvinyl alcohol?PVA?were selected as immobilized materials because of their good biocompatibility.Immobilization of whole cell of Penicillium purpurogenum Li-3 produced SA/PVA liquid core microencapsulated cells by embedding method.The effect of anions on the catalytic properties will be explored by adding synthetic grafted ionic liquids to the immobilization system.Results:1.The catalytic activity of the whole cell of Penicillium purpurogenum Li-3 was higher than that of the organic ion salt system containing[BMIM]Cl and MBPy Cl in the medium system containing Na PF6and Na Tf2N organic ion salts,the results were significantly different?P<0.05?.The organic ion salts Na PF6 and Na Tf2N have good biocompatibility,of which Na Tf2N organic ion salt is the most significant?P<0.05?,and the sugar metabolism activity of Penicillium chrysogenum whole cells is certainly suppressed due to the organic ion salt medium[BMIM]The effects of Cl and MBPy Cl were most significant in[BMIM]Cl?P<0.05?.The morphology of Penicillium chrysogenum whole cells is less affected by the remaining three organic ion salts except[BMIM]Cl ion salt.The three organic ion salts Na PF6,Na Tf2N and MBPy Cl all have the effect of improving the permeability of cell membranes,of which Na Tf2N has the strongest effect?P<0.05?.2.The structure of the grafted ionic liquid was characterized by FTIR,1H NMR,19F NMR,TG,TEM and EDS.The Impurity contents of chloride ion?Cl-?and lithium ion?Li+?in the grafted ionic liquid are quantitatively detected by IC and ICP-AES.It is proved that the structure of the synthesized product is consistent with the literature and has less impurities.3.The optimal preparation conditions for immobilized cells are based on the diameter,mechanical strength,and catalytic properties of SA/PVA liquid core microencapsulated cells as research indicators to study the impact of various factors on their performance:the SA concentration is 1.0%,the PVA concentration is 6.0%,the infusion solution Ca Cl2concentration is 2.0%,the solidification solution Ca Cl2concentration is 2.0%,the added bacteria amount is 5.0%,and placed in a refrigerator at-18?for 4 h.The mass transfer model was established to calculate the mass transfer coefficient k=0.0741 of GL in SA/PVA liquid core microencapsulated cells.SA/PVA liquid core microencapsulated cells can still maintain 62.02%of the activity after 19 consecutive cultures using shaker culture.The synthetic grafted ionic liquid was added to the immobilization system.The maximum yield of GAMG was when the concentration of Si O2-MIM-Tf2N was 10 m M and Si O2-MIM-PF6was 20 m M.Increased yield compared to microencapsulated cells without added grafted ionic liquid.Conclusion:1.The main factor of biocatalytic activity may be the influence of organic anions?PF6-,Tf2N-?.Ionic liquids with corresponding anions of PF6-and Tf2N-are more suitable as catalytic reaction media for Penicillium purpurogenum Li-3.2.The nano-silica grafted ionic liquid was successfully synthesized,which provided a material basis for further exploration and efficient use of the organic anions PF6-and Tf2N-to promote the biocatalytic system.3.The whole cell immobilization of Penicillium chrysogenum uses SA/PVA liquid core microencapsulation technology,and the stability and reutilization rate of the whole cell can be significantly improved.Then,the nano-silica grafted ionic liquid was embedded in biological microcapsules,which effectively played the role of organic anions PF6-and Tf2N-in promoting the biological catalytic system.
