OBJECTIVE:The prognosis of colorectal cancer is related to the diagnosis stage.The 5-year survival rate is 90% in early diagnosis and the distant metastasis rate is less than 10%.Selecting a "personalized" treatment strategy and providing a prognostic marker for colorectal cancer for early diagnosis provides early diagnosis and prognosis for the development of colorectal cancer through different biomarkers,with the potential to improve the outcome of the disease.METHODS:The colorectal cancer microarray data were downloaded from the GEO database,and the bioinformatics analysis of colorectal cancer gene microarray data was performed using R language.The differentially expressed genes related to colorectal cancer were selected from the existing microarray chip data.The expressed gene was subjected to weighted network co-expression analysis(WGCNA)to identify the gene modules related to the development of colorectal cancer,KEGG and GO enrichment analysis of selected gene modules,and then Cytoscape software visualization module gene co-expression network to find out Core node gene,and Using UALCAN,based on the TCGA database,the survival analysis of core node genes.Finally,the mi RIP database was used to predict the mi RNA of the hub gene,combined with survival analysis to screen mi RNA biomarkers for poor prognosis of colorectal cancer.RESULTS:Analysis of differentially expressed genes by weighted gene co-expression network,low expression genes DHRS11,UGP2,GUCA2 B,TRPM6,MT1 M,high expression genes CMTM2,KIF18 A,NCAPH,BUB1,CEP55,CDC45.Then,the results were verified by TCGA-COAD,and the results were consistent with the analysis.Finally,the survival prognosis analysis of 11 hubgenes based on TCGA-COAD clinical data showed that BUB1,DHRS11 were associated with poor prognosis of colorectal cancer.The mi RNA prediction of BUB1 and DHRS11 using the mir DIP database predicted a total of 15 mi RNAs,of which 3 mi RNA's were associated with poor prognosis of colorectal cancer,hsa-mi R-495-3p,hsa-mi R-125b-5p and hsa-mi R-491-5p.CONCLUSION:Genes BUB1,DHRS11,hsa-mi R-495-3p,hsa-mi R-125b-5p and hsa-mi R-491-5p are associated with poor prognosis of Colorectal cancer.
