| Objectives:The objective was to observe the correlation between Aurora A and the efficacy of immunocheckpoint inhibitor in the treatment of NSCLC,and to further explore the possible mechanisms of high expression of Aurora A in lung cancer cells and immunocheckpoint inhibitor resistance.Methods:The relationship between the expression of Aurora A and the efficacy of immunotherapy in 53 lung cancer patients was detected by immunohistochemistry.m RNA and protein expression of Aurora A in lung cancer tissues with different therapeutic effects were verified by RT-PCR and Western blotting.Western blotting confirmed the correlation between Aurora A and PD-L1 expression.The co-culture model verified the clinical phenomenon.The expression of multiple tumor immune mechanisms associated with Aurora A was observed at the RNA level.Results:1.The expression of Aurora A in NSCLC tissues is associated with the efficacy of immunocheckpoint inhibitors.2.The protein and m RNA expression of Aurora A in NSCLC tissues were verified to be consistent with the results of immunohistochemistry.3.Aurora A is related to the expression state of PD-L1.4.Aurora A may be involved in regulating the effects of immunocheckpoint inhibitors on T lymphocyte killing tumor cells.5.The expression of various tumor immune-related molecules is associated with Aurora A.6.Lung cancer cells with high expression of Aurora A are resistant to Pembrolizumab.Conclusion:The high expression of Aurora A indicates poor clinical prognosis of NSCLCand is associated with drug resistance at immune checkpoint inhibitors,which can be used as A new tumor therapy predictive molecule. |
